ABSTRACT: This study aimed to explore the regulatory effect of sulbutiamine on the gene expression profile of human pancreatic cancer PANC1 cells. PANC1 cells cultured in vitro were treated with 0.18 mM sulbutiamine for 48 hours, and untreated PANC1 cells were set as the control group. Total RNA was extracted from cells of each group after treatment, and high-throughput RNA sequencing (RNA-seq) was performed to detect transcriptomic expression changes and screen differentially expressed genes (DEGs). Subsequently, bioinformatics analyses were conducted to analyze the functional enrichment, signaling pathways and protein interaction networks of the identified DEGs, so as to clarify the transcriptomic characteristics of PANC1 cells in response to sulbutiamine intervention. This study reveals the molecular regulatory mechanism of sulbutiamine against pancreatic cancer at the transcriptome level, providing comprehensive transcriptomic experimental evidence for exploring potential therapeutic targets and signaling pathways of sulbutiamine in pancreatic cancer, and laying a theoretical foundation for the research and development of novel adjuvant therapeutic agents for pancreatic cancer.
