Bulk RNA-seq of peripheral organs from naïve, tBMT, and Mr BMT mice under homeostatic and inflammatory conditions


ABSTRACT: Microglia replacement by bone marrow transplantation (Mr BMT) is an emerging strategy that enables efficient engraftment of donor-derived microglia in the central nervous system. Because the conditioning regimens used for Mr BMT, including myeloablation and CSF1R inhibitor-mediated macrophage depletion, may also affect peripheral tissues, we systematically investigated the long-term impact of transplantation on multiple organs. Bulk RNA-seq was performed on lung, liver, spleen, and kidney tissues from naïve, traditional bone marrow transplantation (tBMT), and Mr BMT mice at 3.5 months of age, as well as from 11.5-month-old naïve and Mr BMT mice with or without LPS challenge. These datasets provide a comprehensive resource for evaluating tissue-specific transcriptional remodeling, donor-derived macrophage adaptation, and maintenance of peripheral immune homeostasis following bone marrow transplantation-based macrophage replacement.
