M6Am-seq profiling of macrophage-specific Pcif1 knockout mice during experimental colitis


ABSTRACT: Inflammatory bowel disease (IBD) is characterized by chronic intestinal inflammation accompanied by alterations in immune cell function and enteric nervous system homeostasis. To investigate the role of PCIF1-mediated N6,2′-O-dimethyladenosine (m6Am) RNA methylation in macrophages during colitis, we generated macrophage-specific Pcif1 knockout mice and performed transcriptome-wide RNA sequencing (RNA-seq) and m6Am profiling (m6Am-seq). These datasets were generated to characterize m6Am-regulated gene expression programs and identify molecular pathways associated with extracellular matrix remodeling, macrophage function, and enteric neuronal regulation during intestinal inflammation.
