Effect on flg22 treatment on Col-0, hir2-2, and hir2-5 seedlings

ABSTRACT: We here analyze the response of Col-0, hir2-2, and hir2-5 seedlings to flg22. HIR2 was shown to interact with PAMP-involved receptor-like kinases and signalling proteins. Furthermore, hir2 mutants are alterred in their ROS response upon PAMP treatment.

Comparison of the response of Col-0 and ate1 ate2 N-degron mutant to flg22

Project description:Arabidopsis mutants of the N-degron pathway have been shown to be either susceptible or resistant to a wide range of pathogens. This includes mutants for the arginine-transferases (ate1-2 ate2-1) and for the downstream E3 ubiquitin ligase PRT6 (prt6). In addition, ate1-2 ate2-1 mutants have been shown to have dampened gene expression changes upon inoculation with Pseudomonas syringae AvrRpm1. Here, we tested whether ate1-2 ate2-1 mutants were impaired for pattern-triggered immunity (PTI) and determined gene expression changes of wild-type Col-0 and of ate1-2 ate2-1 mutant seedlings in the absence/pressence of flg22.

Project description:We reported the flg22-triggered immune responses in roots affect the iron deficiency responses and may link to the function of FLS2 and IMA1 in the root. To identify the underlying mechanism of how the root transcriptome profiles respond to +Fe, +Fe+flg22, -Fe, -Fe+flg22 respectively, and if the flg22 responses is dependent on the function of FLS2 and IMA1 in the root, we performed an mRNA-seq experiments in Col-0, fls2 and UBQ10::mCitrine-IMA1 with different treatments. The differentially expressed genes in response to +Fe, +Fe+flg22, -Fe, -Fe+flg22 were analyzed. It has 36 samples in total, with 3 biogical replicates for each condition and each genotype.

RNA-Seq of Arabidopsis Col-0 and sid2 seedlings treated with L-Glu and urea

Project description:Plant resistance inducers (PRIs) are compounds that protect crops from diseases by activating plant immunity. The exogenous treatment with glutamate (Glu), an important amino acid for living organisms, was shown to induce resistance against fungal pathogen in rice and tomato. To understand the molecular mechanism of Glu-induced immunity, we developed a model system using Arabidopsis thaliana. Here, we found that exogenous treatment with Glu to Arabidopsis enhances resistance against Pseudomonas syringae pv. tomato DC3000 and Colletotrichum higginsianum. Consistently, transcriptome analyses of Arabidopsis seedlings treated with Glu showed that Glu significantly induces the expression of wound, defense, and stress related genes. Interestingly, Glu activates the expression of pathogen or damage associated molecular patterns (PAMP or DAMP)–inducible genes at much later time points than PAMP/DAMPs normally do. Moreover, expression of Glu-inducible genes does not require known components of PAMP receptor complex, glutamate receptors, salicylic acid-biosynthesis enzyme, or glutamate decarboxylase. In addition, Glu also enhances PAMP-inducible immune responses, such as production of reactive oxygen species and mitogen-activated protein kinase activation. These results show that Glu activates PAMP/DAMP-triggered immunity signaling pathway in a novel manner.
