Protocol for scalable generation of uniform 3D porcine oviduct epithelial cell spheroids

ABSTRACT: While previous studies have reported in vitro formation of oviduct epithelial spheroid models in porcine species (sus scrofa), these approaches often rely on manual selection of the formed aggregates. Here, we establish a scalable and reproducible system for generating oviduct spheroids. We describe steps for the isolation of primary secretory and ciliated oviduct epithelial cells and their assembly into spheroids using the AggreWell platform. This approach enables the mass production of uniformly sized spheroids ready for use in experiments such as sperm-oviduct co-incubation experiments.

Project description:Liver fibrosis (LF), driven primarily by activated hepatic stellate cells (aHSCs), progresses to life-threatening end-stage diseases such as cirrhosis, hepatocellular carcinoma. As the only effective treatment, liver transplantation remains limited by donor scarcity. While mesenchymal stem cells (MSCs) exert anti-fibrotic effects via immunomodulation, autophagy regulation, and tissue regeneration, their clinical application is hindered by poor survival, short residence time, and inefficient homing after systemic administration. Recent advances indicate that 3D MSCs spheroids enhance cell viability and paracrine function compared to monodispersed cells. Human umbilical cord MSCs (HUMSCs) are ideal due to low immunogenicity and minimal ethical concerns. Injectable decellularized extracellular matrix (dECM) hydrogels, particularly liver-derived ECM (LdECM), offer a bioactive microenvironment to support cell retention and sustained release.HUMSCs spheroids (HUMSCs sph) were engineered using AggreWell™ plates. An injectable hydrogel was fabricated from decellularized Sprague-Dawley (SD) rat liver scaffolds (LdECM). A novel therapeutic strategy for LF was developed by encapsulating HUMSCs sph within LdECM hydrogels. The structure and characteristics of HUMSCs sph/sus and LdECM were assessed. The efficacy of localized high-density HUMSCs sph delivery was evaluated in a C57BL/6 mouse model of bile duct ligation/carbon tetrachloride (BDL/CCL4)-induced LF, comparing outcomes against equivalent doses of HUMSCs suspension (HUMSCs sus). HSCs autophagy and activation were detected while co-cultured with HUMSCs sph/sus.

Project description:Our results indicated that conditional knockout of Cdc42 in the epithelial cells of the oviduct isthmus led to defective embryo transport, resulting in blastocytes retention in oviduct, and contributing to the pregnancy failure in PgrCreCdc42f/f mice. To further determine the role of CDC42 in oviduct, RNA-seq analysis was applied in the Cdc42f/f and PgrCreCdc42f/f mice oviduct on day 2 of pregnancy.

Comparison of small medium and large healthy ovarian pig follicles

Project description:Three different stages of pig antral follicles have been studied in a granulosa-cell transcriptome analysis on nylon microarrays (1152 clones). The data have been generated from 7 RNA follicle pools and several technical replicates were made. Four Large, one Medium and two Small follicles pools were considered. For each follicle pool, 2 radioactive labellings were performed. Each membrane was exposed 16 hours (to avoid saturation of the signal of highly expressed genes) and 28 hours (to get some signal from lowly expressed genes). Each probe was hybridised on GPL3971 scag_scai Sus scrofa 1.2K mono array and on GPL3970 scag_scai Sus scrofa 4.6K triplicate array (except for GFS2172 which was labelled only once and hybridised onto 2 GPL3971 scag_scai Sus scrofa 1.2K mono array membranes), so that 4 spots are available for each gene (and 2 spots for GFS2172), for a given RNA and a given radioactive labelling. Keywords: granulosa-cell transcriptome analysis data consisted in (6 RNA x 2 labellings) + (GFS2172 RNA X 1 labelling)= 13 probes, 26 hybridisations, 52 images.

Total RNA sequencing in multiple Sus Scrofa tissues reveals novel long non-coding RNAs functioning in skeletal muscle development

Project description:Long non-coding RNAs (lncRNAs) play important roles in diverse biological processes. However, the landscape of lncRNAs is largely unclear in Sus scrofa. Here we performed stranded RNA-seq on total RNA libraries from over 100 samples of Sus scrofa tissues. We identified 10,813 lncRNAs in Sus scrofa, of which 9,075 are novel. 57% of these lncRNAs were conserved in both human and mouse. These conserved lncRNAs tend to be more tissue-specific than pig-specific lncRNAs, and enriched in reproducible organs (i.e. testis and ovary). We characterized a group of lncRNAs potentially involved in the skeletal muscle development. One such lncRNA, a homolog of maternally expressed gene 3 (MEG3), was specifically expressed in the skeletal muscle at early developmental stage. And its expression pattern is conserved in pig and mouse. By over-expressing and knocking down MEG3 in mouse myoblast cell lines, we demonstrated its novel function as a myoblast proliferation suppressor.

The equine embryo influences immune related gene expression in the oviduct

Project description:While the equine oviduct clearly affects early embryo development and while the selective transport of equine embryos through the oviduct indicates a reciprocal interaction, the influence of the embryo on gene expression in the oviduct remains to be determined in the horse. The aim of this study was to examine this by means of RNA sequencing. Four days after ovulation, the oviduct epithelial cells ipsilateral and contralateral to the ovulation side from five cyclic and five pregnant mares were collected. mRNA was extracted and samples were sequenced using the Illumina Hiseq-2000 sequencer. Data analysis was performed with the CLC Genomics software and differentially expressed genes (DEGs) were determined (p-value ≤ 0.05 and absolute fold change ≥ 2). ClueGO was used for functional interpretation. A total of 26,991 genes was identified and 253 genes were found to be upregulated and 108 to be downregulated in the pregnant ipsilateral oviduct, when compared to the cyclic ipsilateral oviduct. Comparison of the ipsilateral and the contralateral oviduct indicated 164 DEGs in pregnant mares and 77 DEGs in cyclic mares. Functional enrichment analysis only detected differences in the comparison of pregnant and cyclic ipsilateral oviducts and showed that the equine embryo affects the expression of immune response related genes in the oviduct, with marked upregulation of interferon associated genes. This research represents the foundation for further assessment of the role of specific genes in the early embryo-maternal dialogue of the horse.

Mating induces gene expression shift in chicken oviduct

Project description:We compared gene expression in oviduct tissues between unmated (control) and mated hen. As spermatozoa are foreign to the female reproductive tract therefore we were interested to look at how spermatozoa survive in the female reproductive tract and keep their fertilization potentiality. To check that we collected tissues from oviduct of both control and mated female chicken and compared if sperm deposition to the oviduct made any gene expression shift related to sperm survival.
