A database of extrachromosomal circular DNA (ecDNA) profiles across primary intracranial tumors

ABSTRACT: Extrachromosomal circular DNAs (ecDNAs) have recently emerged as critical drivers of tumor evolution, distinct from linear chromosomal DNA. The presence of ecDNA often correlates with poor patient outcomes and rapid cancer progression, suggesting a potential role as a biomarker for early detection. Here, we present a dataset of ecDNA landscapes generated via extrachromosomal circular DNA sequencing (Circle-Seq) across four types of primary intracranial tumors: glioblastoma, meningioma, pituitary adenoma, and vestibular schwannoma. We provide comprehensive functional annotations of the derived ecDNA profiles, characterizing their genomic distribution and potential oncogenic content. This study characterizes the ecDNA population within these distinct brain tumor subtypes, offering a reliable resource for future biomarker identification and tumour-specific mechanistic investigations.
