Myocardial transcriptome of left ventricular tissue from a murine arthritis model (RA+ vs RA-)


ABSTRACT: Rheumatoid arthritis (RA) is associated with an approximately two-fold increased risk of heart failure not explained by ischaemic heart disease, implicating systemic inflammation as a candidate driver of myocardial injury. To test whether sustained RA-associated systemic inflammation alters the myocardium, male BALB/c mice underwent a modified delayed-type hypersensitivity arthritis (DTHA) protocol with weekly re-induction over eight weeks (RA+), while control animals received a vehicle injection protocol (RA-). Bulk RNA sequencing was performed on left ventricular myocardium collected at the end of the study, to characterise the myocardial transcriptional response to sustained systemic inflammation in hearts showing no overt structural or functional remodelling by cardiac MRI, histology, or conventional biomarkers.
