The relationship between cell surface CD11c and neutrophil transcriptomic profiles

ABSTRACT: CD11c is traditionally considered as a dendritic cell marker, but we and others reported that it is expressed in other type of cells. We recently reported that CD11c was expressed in the intracellular space of neutrophils and delineated its function. However, we do not know any functional role of cell surface CD11c in neutrophils. We decided to study the role of cell surface CD11c by sorting into CD11c negative/low neutrophils and CD11c high neutrophils.

Transcriptome analysis of myeloid cells (neutrophils, macrophages and dendritic cells) isolated from the spinal cord of mice with experimental autoimmune encephalomyelitis (EAE)

Project description:We have discovered that neutrophils that infiltrate the spinal cord of mice with EAE, a model of multiple sclerosis, but not intravascular neutrophils that crawl on the luminal endothelial surface, bear on their surface the adhesion molecule Icam1. The goal of this experiment was to use Icam1 to isolate these two neutrophil subpopulations in order to compare their transcriptomes and gain insights into their properties. To this end, EAE was induced in C57BL/6J mice by immunization with myelin oligodendrocyte glycoprotein (MOG) peptide (a.a. 35-55) and adjuvants (i.e. complete Freund’s adjuvant and pertussis toxin). On day 15 post-immunization, intravascular neutrophils (CD45hiCD11b+CD11c−Ly6g+Icam1−) and extravasated neutrophils (CD45hiCD11b+CD11c−Ly6g+Icam1+) were isolated from spinal cords by FACS. For comparison, we simultaneously isolated two other populations of myeloid cells: macrophages (CD45hiCD11b+CD11c−Ly6g−) and dendritic cells (CD45hiCD11b+CD11c+Ly6g−). RNA was analyzed in biological duplicate using Affymetrix GeneChip Mouse Gene 2.0 ST arrays.

Project description:We previously reported that a synthetic Nod1 ligand, FK565, induced coronary arteritis in mice similar to Kawasaki disease. However, the molecular mechanisms underlying this site-specific inflammation have remained elusive. In this study, we found that CD11c+MHC class II+ cells accumulated in the heart of FK565-treated mice prior to arteritis development. We used microarray analysis to detail gene expression of CD11c+MHC class II+ cells. To obtain gene expression profile of CD11c+MHC class II+ cells, we isolated these cells from hearts of FK565-treated mice. Briefly, female mice at 8weeks age were administered 500 μg of FK565 subcutaneously at day 0 and day 3. At day6, murine hearts were removed and digested with collagenase. CD11c+MHC II+ cells were sorted as PI–CD45+Ly6G–NK1.1–CD11b+CD11c+MHC II+ using FACS Aria cell sorter (BD Biosciences). Sorted cells were subjected to RNA preparation. Two independent replicates from ten mice were made.

CD11c-expressing microglia are transient, driven by interactions with apoptotic cells

Project description:Microglia, the parenchymal macrophage of the central nervous system serve crucial remodeling functions throughout development in response to developmental cues. Microglia are transcriptionally heterogenous, suggesting that distinct microglial states confer distinct roles. Currently, little is known about how dynamic these states are, the cues that promote them, or how they impact microglial function. In the developing retina, we previously found a significant proportion of microglia that express CD11c (Integrin αX, complement receptor 4, Itgax). which has also been reported in other developmental and disease contexts. Here, we sought to understand the regulation and function of CD11c+ microglia by examining the developmental contexts and dynamics for the emergence of CD11c+ microglia. We find that microglial CD11c expression tracks with prominent waves of neuronal apoptosis in postnatal retina. Using genetic fate mapping, we provide evidence that microglia transition out of the CD11c state to return to homeostasis. We show that CD11c+ microglia have elevated lysosomal content and contribute to the clearance of apoptotic neurons, and found that acquisition of CD11c expression is, in part, dependent upon the TAM receptor Axl. Using selective ablation, we find similar effects on apoptotic cell clearance with depletion of CD11c+ versus non-CD11c microglia, suggesting that CD11c+ microglia are not uniquely critical for phagocytic clearance. Together, our data suggests CD11c+ microglia are a transient state induced in response to developmental apoptosis rather than a specialized subset mediating phagocytic elimination.
