Host transcriptional signatures associated with disease tolerance and environmental persistence in a mosquito-microsporidian system

ABSTRACT: Experimental evolution and RNA sequencing were combined in the mosquito-microsporidian system Anopheles gambiae-Vavraia culicis to test whether parasite lineages selected for early or late transmission differ in the host transcriptional responses they elicit. Mosquitoes were infected with evolved parasite lineages and sampled at a common sporulating stage. Co-expression analyses identified host gene modules associated with infection background, virulence and environmental persistence.

Host complement C3 promotes malaria transmission by killing symbiotic bacteria in the mosquito midgut

Project description:Host-derived factors are sucked into midgut of mosquitoes during natural malaria transmission, but their influence on malaria transmission is largely unknown. We reported that mouse complement C3 taken into mosquitoes significantly promoted malaria transmission either in laboratory or in field. This effect was attributed to the reduction of microbiota abundance in mosquito midgut by host-derived C3 through direct lyses the predominant symbiont bacteria Elizabethkingia anopheles. Elizabethkingia anopheles symbiont bacteria were demonstrated to be detrimental to malaria sexual stages in mosquitoes. Strikingly, the promoted effect of host C3 on malaria transmission was confirmed by laboratory mosquitoes membrane-feeding on Plasmodium falciparum. Therefore, we reveal a novel strategy of malaria parasite to utilize host complement C3 to promote its transmission, and the administration of C3 inhibitor would provide us a novel strategy to control malaria transmission.

Project description:In this study we determine the effects of silencing adiponectin receptor in A. gambiae mosquitoes after taking Plasmodium gambiae infected blood meals. dsRNA (dsAdpR and dsGluc) microinjected A. gambiae mosquitoes were fed on P. berghei-infected Swiss Webster mice. Two days after a blood meal, the mosquitoes were collected for midgut dissection.

Project description:The Anopheles gambiae midgut harbors bacteria that proliferate upon a blood feed. We used microarrays to examine the midgut gene expression response at early stages (3hours) after an artifitial meal containing heat killed bacteria. Anopheles gambiae G3 mosquitoes 5-6 day-old were fed BSA (20% in PBS with fresh 10 mM sodium bicarbonate) with or without heat killed E. coli (equivalent of 2.5 ml of 0.8 OD) . Three pools of 10 mosquito midguts were dissected after 3h and processed for microarray analysis of gene expression.

Transcriptome of developing testes from the African malaria mosquito Anopheles gambiae

Project description:Spermatogenesis is a highly complex developmental process, during which diploid germline stem cells are transformed into motile haploid spermatozoa. The process involves a precisely regulated action of a large number of genes, making the testes the most distinct tissue within the body. Testis transcriptome has been analysed in several groups of animals, but never systematically studied in mosquitoes. This dataset, consisting of the transcriptome of the developing testes from late larvae and early pupae of the African malaria mosquito, Anopheles gambiae, closes the existing gap.

The mosquito transcriptome is shaped by the individual and interactive effects of warmer temperature and aging

Project description:In mosquitoes, warmer temperature accelerates the progression of senescence, weakening their ability to survive and respond to an infection. Thus, we hypothesized that this phenomenon is driven by changes in gene expression. To test this hypothesis, we employed an RNA-sequencing approach to investigate how warmer temperature and aging, individually and interactively, shape the transcriptome of the female African malaria mosquito, Anopheles gambiae. Warmer temperature increases the expression of genes involved in metabolism and cell signaling but decreases the expression of genes involved in replication, transcription, translation, and protein processing. Aging decreases the expression of metabolism-related genes but increases the expression of protein processing and transcriptional genes. However, early and late aging have opposing effects on many biological processes, with 1-day-old mosquitoes having a largely different transcriptomic profile than older mosquitoes. Importantly, warmer temperature and aging interactively shape gene expression patterns, where, for example, aging-dependent changes in immune gene expression accelerate at warmer temperatures. Altogether, warmer temperature and aging, individually and interactively, alter the mosquito’s transcriptomic profile, reflecting changes in their ability to survive and overcome an infection.

CpG Oligodeoxynucleotides treatment of Anopheles mosquitoes

Project description:In the present study, we have investigated the effect of CpG Oligodeoxynucleotides (CpG-ODN) on the outcome of Plasmodium infection of the mosquito vectors Anopheles stephensi and Anopheles gambiae and on the modulation of mosquito immunity to Plasmodium. Anopheles mosquitoes inoculated with CpG-ODN showed significant reduction of Plasmodium infection rate and intensity. Microarrays were used to profile transcription of fat-body from CpG-ODN-treated mosquitoes. Mosquitoes were dissected 18h after ODN inoculation (immediately before feeding). Batches of 20 to 30 fat bodies (abdomen without midgut, ovaries and malpighian tubule]) were dissected in cold DEPC-treated phosphate-buffered saline (PBS) and processed for RNA preparation. Mosquitoes treated with CpG-ODNs are less susceptible to Plasmodium infection. Transcription profile of fat body indicates that protection was associated with coagulation/wound healing, while melanization appears to be depressed.
