Transcriptomic profiling of RAW 264.7 macrophages treated with IL-4 and CGRP versus IL-4 alone

ABSTRACT: Sensory nerve–immune crosstalk has emerged as a critical regulator of tissue repair, yet the molecular mechanisms by which sensory neurons communicate with macrophages to influence bone regeneration remain incompletely understood. In particular, whether calcitonin gene related peptide (CGRP), a neuropeptide released from sensory neurons, directly modulates macrophage transcriptional programs to promote a pro-healing phenotype has not been systematically characterized. Here, we performed RNA sequencing on RAW 264.7 murine macrophages stimulated with IL-4 alone (control) or IL-4 combined with recombinant CGRP (experimental) for 24 hours. We identified a total of 169 differentially expressed genes (|log₂FC| > 0.585, p < 0.05) between the CGRP treated and control groups, with significant enrichment in inflammation and immune response related pathways. Among the upregulated genes, we identified Hspb1, a small heat shock protein previously implicated in cytoskeletal dynamics and stress responses, as a key downstream mediator of CGRP induced M2 macrophage polarization. This dataset provides a valuable transcriptomic resource for understanding how sensory neuron derived CGRP reprograms macrophage gene expression to facilitate M2 polarization, and may help uncover novel molecular targets for harnessing neuro-immune interactions to enhance bone regeneration.
