PAR-CLIP-sequencing of PC9 lung cancer cells to identify FUS bingind sites

ABSTRACT: The prognosis of non-small cell lung cancer (NSCLC) with leptomeningeal metastases (LM) is dismal. The regulators of LM progression remain elusive, thus impeding effective clinical intervention. Here, we discovered that Fused in sarcoma (FUS) ablation promoted LM in lung cell lines. As FUS is a multifaceted DNA-/RNA-binding protein involved in transcription, we explored how it represses CD36 with PAR-CLIP-seq (photoactivatable–ribonucleoside-enhanced cross-linking and immunoprecipitation, followed by high-throughput sequencing) in PC9 cells.
