Single-cell transcriptomic analysis of Wfdc2-deficient mouse lungs and response to amiloride treatment


ABSTRACT: This study investigated the transcriptomic consequences of WFDC2 deficiency in mouse lungs and the response to ENaC inhibition. WFDC2 deficiency caused respiratory distress associated with epithelial defects, immune activation, and hyperactivation of the epithelial sodium channel (ENaC). Single-cell RNA sequencing was used to profile lung cells from wild-type, Wfdc2 knockout, Wfdc2 p.C147W knock-in, and amiloride-treated mice. The data reveal altered epithelial cell differentiation and increased immune cell populations in Wfdc2-deficient lungs, while amiloride treatment partially restored epithelial cell states and respiratory phenotypes.
