Project description:Skin serves as the primary barrier against hazardous substances, and timely wound closure is an essential initial step in wound regeneration. However, intrinsic skin regenerative capacity is severely limited in critical-size acute wounds due to insufficient cell migration and tissue regeneration. Electrical stimulation restores endogenous wound electric fields and promotes cell migration and proliferation, thereby accelerating wound regeneration. Repetitive stimulation with identical electrical waveforms may attenuate electrophysiological responsiveness. Here, we present a miniaturized, lightweight, inertia-driven triboelectric nanogenerator–based electroceutical platform (2 cm3, 4.9 g) that enables self-powered operation with bioresorbable electrodes. In vitro studies demonstrate that electric fields and charges enhance cell migration and exosome secretion. In vivo animal studies confirm that motion-modulated, electrical waveforms delivered to the wound bed promote angiogenesis and epidermal maturation and stabilize the wound-associated microbiome. Collectively, these findings establish an electroceutical strategy for effective wound management in aging and impaired healing environments.

Project description:We report a high resolution catalouge of NMD substrates using RNA-Seq. We discovered several hundred new substrates for NMD. Using published ribosome footprint profiling data, we measured ribosome densities of normal-looking NMD substrates and non-NMD substrates. NMD substrates exhibited a striking difference in normalized ribosome occupancy in wild-type and UPF1 cells. We also found that normal looking NMD substrates have higher ratio of out of frame reads, lower codon optimalites and a higher propensity to have long stretches of non-optimal codons.

Project description:Tissue response following implantation determines the success of the healing process. This response is not only dependent on the chemical properties of the implant surface but also by the surface topography or its roughness. Although in vitro and in vivo studies show improved results with rough- and fluoride-modified implants, the mechanisms behind these findings are still unknown. Here, we have used a two step procedure to identify novel genes related to the early cell response of primary human osteoblasts to roughness and fluoride-modified titanium implants. 217 genes were identified by microarray analysis as response genes to roughness and 198 genes as response genes to fluoride. 11 of these identified genes have been related to bone and mineralization and were further investigated by real-time RT-PCR. After one day of culture, TLR3, ANKH, DCN, OC and RUNX2 were classified as responsive genes to roughness; DLX2 and TUFT1 as responsive genes to fluoride treatment. COLL-I, PTHLH, HES1, FST, ENPP1 and THRA as responsive genes to both, roughness and fluoride treatment. In conclusion, our strategy was useful for identifying novel genes that might be involved in the early response of osteoblasts to roughness and fluoride treatment of titanium implants. Human osteoblasts were cultured for 24 hours on titanium disks modified either with polished surfaces (P), grit-blasted surfaces (GB), or grit-blasted High Fluor treated surfaces (GB-HF). After 24 hours of culture RNA was isolated. 3 arrays were hybridized, one with the isolated RNA from cells cultured on titanium polished surfaces (P), one with the isolated RNA from cells cultured on titanium grit-blasted surfaces (GB) and one with the isolated RNA from cells cultured on titanium grit-blasted HF treated surfaces (GB-HF).

Project description:Study on changes in gene expression in primary cultures of neonatal rat ventricular cardiomyocytes to electric stimulation. Through comparing non-stimulated, stimulated and blebbistatin supplemented and stimulated cultures we set out to identify the genes that are specifically activated by electric pulsing separate from cardiomyocyte contractions. After initial recovery phase, primary cultures of neonatal rat ventricular cardiomyocytes were cultured for 3 days without pulsing, with electric pulsing or with electric pulsing combined with blebbistatin. Per treatment: 3 arrays, with samples obtained from 3 separate series of cardiomyocyte isolation and culturing. Per array: sample prepared from pooled (1:1) RNA from duplicate experiments.

In this experiment, human bone marrow mesenchymal stem cells were seeded on substates with different ECM rigidity, and the changes of gene expression were detected by chips, so as to explore the effect of extracellular microenvironment on stem cell differentiation.
