Bulk RNA seq data of transcription factor ELF-1 in mouse BMDM derived macrophages.

ABSTRACT: To define the functional impact of ELF-1 on macrophage-driven AKI, we induce M1 polarization in ELF-1-deficient murine macrophages and perform RNA-seq. Building on clinical correlations between ELF-1 and inflammatory cytokines, this study systematically profiles ELF-1-dependent genes and pathways, with the goal of elucidating how ELF-1 orchestrates M1 polarization and contributes to the pathogenesis of AKI following renal transplantation.

Project description:Acute kidney injury (AKI) is an unavoidable complication following renal transplantation and a critical determinant of patient prognosis. M1-polarized macrophages are known to exacerbate the progression of AKI. Preliminary clinical data reveal a positive correlation between the expression level of E74-like ETS transcription factor 1 (ELF-1) in macrophages from post-transplant patients and the levels of pro-inflammatory cytokines. However, the role of ELF-1 in M1 macrophage polarization and the pathogenesis of AKI remains unclear. In this study, we aim to investigate the role of ELF-1 in M1 macrophage polarization and AKI pathogenesis by employing ELF-1-deficient murine macrophages and performing ChIP-seq to identify downstream genes regulated by ELF-1.

DNA methylation analysis during neutrophilic granulopoiesis under ELF-MF exposure

Project description:To address whether ELF-MF influences the epigenetic landscape in differentiating haematopoietic cells, we performed an in vitro haematopoietic differentiation under powerline-simulating ELF-MF exposure (50 Hz, 1 mT, 5’ on/10’ off) in comparison to sham exposure (50 Hz, 7 µT, 5’ on/10’ off) or no exposure. We differentiated CD34+ cells (allcells, CB008F, lot: CBC121009M) isolated from human cord blood into the neutrophilic lineage, and performed genome-wide methylation analysis at single CpG sites of CD34+ cells and day 5 progenitors, using the Illumina Infinium HumanMethylation 450 platform. Exposure to ELF MF did not significantly impact the programming of DNA methylation pattern during granulopoiesis. However, ELF MF exposure has an influence on the robustness of the epigenetic landscape during global chromatin programming of granulopoiesis. Our data suggests that ELF MF has a stochastic effect on the epigenetic landscape of individual cells.

Genome-wide analysis of histone modifications in Jurkat cells upon ELF-MF exposure

Project description:To address whether ELF-MF influences the epigenetic landscape in the T cell lymphoma cell line Jurkat , we exposed Jurkat cells in the exponential growth phase to ELF MF or a sham control for 72 h, applying intermittently (5’ on/10’ off) a 50 Hz sine ELF-MF at a flux density of 1 mT. Additionally, cells were treated with the histone deacetylation inhibitor trichostatin A (TSA) at a subtoxic dose of 10 nM. Genome-wide profiling of active and repressive histone modifications did not reveal significant alterations. However, ELF-MF exposure has an influence on the robustness of the epigenetic landscape in leukaemic cells. Our data suggests that ELF-MF has a stochastic effect on the epigenetic landscape of individual cells.

Transcription factor ELF-1 protects against colitis by maintaining intestinal epithelium homeostasis

Project description:Inflammatory bowel disease (IBD) is a chronic, relapsing, and remitting disease characterized by chronic inflammation in the gastrointestinal tract. The exact etiology and pathogenesis of IBD remain elusive. Although ELF-1 has been known to be highly expressed in epithelial cells for past twenty years, little is known about its function in epithelial cells and epithelial-related IBD. Here, we demonstrated that ELF-1 deficiency in mouse lead to exacerbated DSS-induced colitis, marked by inflammation dominated by neutrophil infiltration and activation of IL-17 signaling pathways in various immune cells, including Th17, ILC3, γδT and NKT cells. Bone marrow transfer experiments confirmed ELF-1 deficiency in non-hematopoietic cells intrinsically worsened DSS-induced colitis. On one hand, ELF-1 deficiency enhanced the production of pro-inflammatory chemokines in colonic epithelial cells, leading to extensive infiltration of neutrophils and other immune cells into the colonic mucosal tissue. On the other hand, ELF-1 directly regulated the expression of the Rack1 gene in colonic epithelial tissue, which has been proved to play critical roles in maintaining intestinal homeostasis. Altogether, ELF-1 plays a protective role in colitis by maintaining intestinal epithelium homeostasis.

MicroRNA expression proï¬ling in GC-2 cells following 50 Hz ELF-EMF

Project description:Aberrant miRNA expression has been related to the development of human germ cell tumors, but little is known about effect of ELF-EMFs on miRNA expression. ELF-EMFs may epigenetically modify cells, which may account for the adverse effects of ELF-EMFs on the male reproductive system. To identify miRNAs that were differentially expressed between the sham and ELF-EMF exposure groups, we performed an Affymetrix microarray analysis to establish the miRNA expression profiles. Mouse spermatocyte-derived GC-2 cells were intermittently exposed to a 50 Hz ELF-EMF for 72 h (5 min on/10 min off) at magnetic field intensities of 1 mT and 3 mT. miRNA expression was profiled using Affymetrix Mouse Genechip miRNA 3.0 arrays.
