CircRNA BRAKE on eWAT after specific knockdown in adipose tissue

ABSTRACT: Mice homozygous for the circRNA BRAKE conditional allele and the Adipo-Cre transgene were generated. Adipose tissue was collected from 8-week-old animals, and primary adipocyte progenitor cells (APCs) were isolated. Following adipogenic differentiation into mature adipocytes, transcriptomic profiling was performed.

Identification of metabolically distinct adipocyte progenitor cells in human adipose tissues

Project description:Adipocyte progenitor cells (APCs) provide the reservoir of regenerative cells to produce new adipocytes, although their identity in humans remains elusive. Using FACS analysis, gene expression profiling and metabolic and proteomic analyses, we identified three APCs subtypes in human white adipose tissues. The APC subtypes are molecularly distinct but possess similar proliferative and adipogenic capacities. Adipocytes derived from APCs with high CD34 expression exhibit exceedingly high rates of lipid flux compared with APCs with low or no CD34 expression, while adipocytes produced from CD34- APCs display beige-like adipocyte properties and a unique endocrine profile. APCs were more abundant in gluteofemoral compared with abdominal subcutaneous and omental adipose tissues, and the distribution of APC subtypes varies between depots and in patients with type 2 diabetes. These findings provide a mechanistic explanation for the heterogeneity of human white adipose tissue and a potential basis for dysregulated adipocyte function in type 2 diabetes.

circRNA Microarray Service for 6 mouse eWAT samples

Project description:We have completed the Arraystar Mouse circRNA Array V2 analysis of the 6 samples that you submitted. Total RNA from each sample was quantified using the NanoDrop ND-1000. The sample preparation and microarray hybridization were performed based on the Arraystar’s standard protocols. Briefly, total RNAs were digested with Rnase R (Epicentre, Inc.) to remove linear RNAs and enrich circular RNAs. Then, the enriched circular RNAs were amplified and transcribed into fluorescent cRNA utilizing a random priming method (Arraystar Super RNA Labeling Kit; Arraystar). The labeled cRNAs were hybridized onto the Arraystar Mouse circRNA V2 Array (8x15K, Arraystar). After having washed the slides, the arrays were scanned by the Agilent Scanner G2505C

Transcription factor PATZ1 promotes adipogenesis by controlling promoter regulatory loci of adipogenic factors [ChIPseq_D-2_D0_D5_D8]

Project description:White adipose tissue (WAT) plays a central role in lipid storage and systemic energy, lipid, and glucose homeostasis. Understanding the intricacies of adipocyte formation could inform therapies for obesity and metabolic disorders. We have identified the POZ/BTB and AT Hook Containing Zinc Finger 1 (PATZ1) protein as an adipogenic transcription factor through an unbiased high-throughput cDNA screen for transcriptional modulators of adipogenesis. PATZ1 is expressed by both human and mouse adipocyte precursor cells (APCs) and adipocytes, and in cell models, PATZ1 expression promotes adipogenesis through a mechanism dependent on protein-protein interaction and DNA binding. Both adipocyte-specific and APC-specific ablation of PATZ1 in mice leads to decreased fat mass and hypertrophied adipocytes. Genome-wide PATZ1 DNA binding analyses using ChIP-Seq suggest PATZ1 facilitates adipogenesis through interactions with transcription factor machinery at the promoter regions of critical early adipogenic factors and histone modifiers. Purification of the PATZ1 complex showed that General Transcription Factor 2I (GTF2I) associates with PATZ1 in a differentiation-dependent manner. Downregulation of GTF2I levels during adipogenesis markedly augments PATZ1 adipogenic function, suggesting a repressive interaction between GTF2I and PATZ1. These findings identify PATZ1 as a regulator of both adiposity and adipocyte differentiation programs and advance our understanding of the complex transcriptional mechanisms underlying adipose tissue development and homeostasis.
