Single-cell RNA-seq of synovial fluid cells from osteoarthritis control patients

ABSTRACT: This dataset contains processed 10x Genomics single-cell RNA-seq data from synovial fluid cells of three treatment-naive osteoarthritis (OA) patients. These OA samples were used as controls in the published Nature Communications study "Single cell immunoprofile of synovial fluid in rheumatoid arthritis with TNF/JAK inhibitor treatment" (doi: 10.1038/s41467-025-57361-0), which profiled synovial fluid immune cells to characterize rheumatoid arthritis-associated cellular states and treatment-related immune remodeling.

Transcriptional analysis of normal and pathological synovial fibroblasts

Project description:The aim of this study was to compare gene expression between two pathological groups of human synovial fibroblasts (SF) from rheumatoid arthritis (RA) and osteoarthritis (OA) synovial tissues with normal SF from healthy individuals (HSF). We used microarray expression profiling in SF cultured from OA, RA and normal synovial tissues. We found larger numbers of transcripts with differential expression in OASF compared to the other groups than in RASF compared to HSF. This data demonstrate that cultured OASF display a more robust transcriptomic profile than RASF when compared to HSF. Synovial fibroblasts were obtained from 9 patients with rheumatoid arthritis (RASF), 11 sex and age matched adult healthy donors (HSF) and 11 sex and age matched patients with OA (OASF). SF were collected under similar subconfluent conditions 24h after serum addition. 31 microarray data were used for determine the statistical significance (p value) of the differences in gene expression.

Indentification of special gene expressions in synovial tissues of rheumatoid arthritis

Project description:Gene expression microarray was applied to discover novel rheumatoid arthritis (RA)-specific gene expressions by comparing the expression profiles of synovial membranes from patients with RA, osteoarthritis (OA) and ankylosing spondylitis (AS). We performed a gene expression microarray analysis of RA synovial membranes and simultaneously compared the expression profile with the profiles of AS and OA synovial membranes. This study was undertaken to investigate the global gene expression profiles in synovial tissues from RA (n=10), OA (n=7), and AS patients (n=5). The Illumina HumanHT-12 v4 Expression BeadChip were used for a complete genome-wide transcript profiling.

miR-548aj-3p and miR-3127-3p suppress RANKL-facilitated inflammatory cytokines and catabolic factor in osteoarthritis and rheumatoid arthritis

Project description:Osteoarthritis (OA) and rheumatoid arthritis (RA) are high prevalent joint diseases in global. The common pathological features include synovial inflammation, swelling, joint destruction, and bone remodelling. Arthritis development is associated with joint inflammation, particularly in inflamed synovial cells. Synovial inflammation, driven by cytokines such as interleukin-1 beta (IL-1β) and interleukin-6 (IL-6), is a crucial factor in arthritis as it contributes to joint destruction. RANKL is a vital factor that is linked to the activity of osteoclasts and the erosion of bone. Increased levels of RANKL related with the proinflammatory cytokines and cartilage degradation enzymes expression, playing a role in the course of arthritis. Medicines for arthritis have been limited by side effects and individual variability in therapeutic response. More effective treatment and drug options are needed to improve disease progression. miRNAs directly modulate gene transcription as potential option for arthritis therapeutic. Our study revealed that RANKL stimulation in OA and RA synovial fibroblasts upregulated the expression of IL-1β, IL-6 (pro-inflammatory cytokines) and MMP-13 (catabolic factor) by suppressing miR-548aj-3p and miR-3127-3p. Administration of miR-548aj-3p and miR-3127-3p mimics substantially inhibited the expression of IL-1β, IL-6 and MMP-13. We propose a potentially efficacious miRNA therapeutic approach for the treatment of arthritis, with a specific focus on OA and RA.

Project description:To investigate the effects of soluble factors produced by synovial CD8 T cells, we stimulated human rheumatoid arthritis (RA) synovial fibroblasts with supernatants from synovial fluid CD8 T cells, blood CD8 T cells, or synovial fluid CD4 T cells stimulated with anti-CD3/CD28 antibody-coated beads. For comparison, we stimulated RA synovial fibroblasts with recombinant TNF or interferon-gamma or T cell supernatants pre-incubated with TNF-blocking antibodies.
