Single-cell RNA sequencing of liver non-parenchymal cells from hepatocyte-specific ADK-overexpressing and control mice


ABSTRACT: Adenosine kinase (ADK) regulates adenosine metabolism and cellular methylation reactions. This study investigated how hepatocyte-specific ADK overexpression affects the transcriptomic states of liver non-parenchymal cells. Liver non-parenchymal cells were isolated from 5-month-old male hepatocyte-specific ADK-overexpressing mice (Hep-ADKTg) and control mice (Hep-ADKWT) maintained on a standard chow diet. Single-cell RNA sequencing was performed using the 10x Genomics Chromium platform to characterize liver cell populations and evaluate transcriptional changes in immune and stromal cells. The analysis showed that hepatocyte ADK overexpression promoted proinflammatory transcriptional changes in liver macrophages and monocytes.
