GFP ChIL-seq mapping of H3mm15-EGFP chromatin incorporation in transgenic mouse testis

ABSTRACT: H3mm15 is a testis-specific histone H3 variant whose genomic distribution has remained undefined. We applied chromatin integration labeling followed by sequencing (ChIL-seq) with an anti-GFP antibody to map H3mm15-EGFP incorporation in testes of transgenic mice expressing the fusion protein under the CAG enhancer/promoter. H3mm15-EGFP signal accumulated preferentially around transcription start sites, and incorporated genes were biased toward the transcriptionally active fraction of the genome. Non-transgenic littermates, which lack the EGFP fusion protein, were processed in parallel as controls.
