Multidimensional Evaluation of UC-MSCs: Comparative Therapeutic Efficacy of Intrathecal and Intravenous Administration in a Rat Traumatic SCI Model

ABSTRACT: Spinal cord injury (SCI) is a disabling neurological disorder with very limited therapeutic strategies. Human umbilical cord derived mesenchymal stem cells (UC MSCs) have shown therapeutic potential for SCI, but the optimal administration route remains undetermined. This study compared the efficacy and mechanisms of GMP grade UC MSCs delivered via intrathecal (MSC i.t) and intravenous (MSC i.v) injection in a rat traumatic SCI model.

A Biodistribution Study of OTV vs. ASO by Intravenous or Intrathecal Administration in Cynomolgus Monkeys

Project description:The objective of this study is to evaluate ASO biodistribution and pharmacodynamic response following intravenous delivery of conjugated OTV, which utilizes transferrin-receptor binding to deliver ASO, vs. intrathecal delivery of naked ASO in cynomolgus monkeys, the current gold standard for ASO delivery. Three groups were included for comparison: 1) Negative control cohort = Four biweekly intravenous doses of saline (n=3); 2) IT cohort = Single intrathecal dose of 4mg MALAT1 ASO (n=3); 3)  OTV multi dose cohort = Four biweekly intravenous doses of 30mpk OTV:MALAT1 (n=3). Tissues were collected two weeks after the final dose to allow sufficient time for target knockdown. Brain, spinal cord, and peripheral tissues were dissected and frozen for molecular analysis. Compared to an intrathecal injection of ASO, systemic administration of OTV resulted in significantly more homogenous biodistribution of ASO in the central nervous system as measured by a probe-based hybridization assay as well as staining with an anti-ASO antibody. One potential consequence of heterogeneous ASO distribution is heterogeneous target knockdown throughout the CNS. We observed more consistent target knockdown throughout the cortex, subcortical areas, and spinal cord of monkeys dosed peripherally with OTV. By contrast, monkeys dosed intrathecally with naked ASO showed much more knockdown in the spinal cord compared to the brian, consistent with ASO distribution.

The initiating IL-10 production dominates the therapy of mesenchymal stem cell scaffold in spinal cord injury

Project description:spinal cord injury (SCI) is an acute damage to central nevous system, resulting in severe morbidity and permanent disability. Locally implanting scaffold system immobilized mesenchymal stem cellshas been widely proven to promote the locomotor function recovery of SCI rat, but the underlying mechanism remains elusive. Here we constructed a hyaluronic acid scaffold system (HA-MSC) to accelerates human MSC adhesive growth and prolongs the survival time of MSC in the lesion of SCI rats. And the residual MSCs regulate the local immune responses by upregulating anti-inflammatory cytokines. Interestingly, the dramatically increased but transient expression of IL-10 is found secreted by MSCs in the first week. Blocking the function of the initiating produced IL-10 by antibody totally abolishes the neurological and behavioral recovery of SCI rats, indicating a core role of IL-10 in SCI therapy of HA-MSC implantation. Transcriptome analyses indicate that IL-10 selectively promotes the migration and cytokine secretion programs of MSC, which in turn help MSC to exert its anti-inflammatory therapeutic effects. Our findings thus highlight a novel role of IL-10 in regulating MSC migration and cytokine secretion and determine the vital role of IL-10 in the domination of MSC treatment for spinal cord repair.

Neuroligin 3 promotes functional recovery of spinal cord injury through coordinating with Sar1a and Hspa8 at synapses

Project description:The recovery of locomotor function following incomplete spinal cord injury (SCI) primarily relies on the precise reconstruction of synaptic communications between spared corticospinal tract (CST) axons and propriospinal neurons. However, components capable of rebuilding synaptic machinery after injury remain elusive. Here, using RNA-Seq of human umbilical cord-derived MSCs (hUC-MSCs) before and after transplantation in SCI rats, along with in vitro and in vivo functional screenings, we identified Neuroligin 3 (NLGN3) as a spinal cord microenvironment (SCE)-activated cell adhesion molecule (CAM) in hUC-MSCs, promoting MSC-mediated functional recovery of SCI. Importantly, spinal neuron-specific activation of Nlgn3 led to significant functional improvement in SCI rats, resembling the effects of hUC-MSC transplantation. We elucidated the protein interactome of Nlgn3, enriched with proteins involved in synaptic transmission. Specifically, Nlgn3 interacted with synaptic vesicle (SV)-associated proteins, Sar1a and Hspa8, modulating their recruitment at synapses. Remarkably, restoring either of these two proteins in injured spinal neurons improved locomotor recovery of SCI, with further enhancement when combined with Nlgn3. Thus, our results not only reveal the previously unknown therapeutic effect of Nlgn3 in SCI repair but also propose a novel approach for treating SCI by targeting protein complexes centered around Nlgn3. This study also suggests that MSCs can serve as a stem cell platform for identifying innovative therapeutic targets and mechanisms to reestablish corticospinal-spinal relay circuits after SCI.

Epigenetic and in vivo comparison of diverse mesenchymal stromal cell sources reveals an endochonrdal signature for human hematopoietic niche formation

Project description:Despite similarities in morphology, phenotype and in vitro behavior, Mesenchymal Stromal Cells (MSC) form various tissue sources show striking differences in their in vivo potential to form bone, cartilage and hematopoietic support tissue. Comparing four commonly use MSC sources (bone marrow (BM), white adipose tissue (WAT), umbilical cord (UC) and skin) we found only bone marrow (BM)-derived MSCs capable of endochondral ossification and marrow attraction. To gain mechanistic insights explaining this differences we analyzed gene expression characteristics of MSC from all four tissue sources using Affymetrix Genechip Human Gene 2.0 ST Array. MSCs from BM, WAT, UC and skin were isolated using plastic adherence. Cells were expanded in standard alpha-MEM supplemented with pooled human platelet lysate fully replacing fetal bovine serum. MSCs were subcutaneously implanted into immune-compromised mice to comparatively evaluate bone formation and subsequent bone marrow attraction. In parallel we have isolated RNA from all sources to analyze tissue source specific gene expression profile.

Effects of human adipose tissue-derived and umbilical cord tissue-derived mesenchymal stem cells in a dextran sulfate sodium-induced mouse model

Project description:Background: Mesenchymal stem cells (MSCs) can be acquired from medical waste. MSCs are easily expanded and have multiple functions, including anti-inflammatory effects. We evaluated the effects of human adipose tissue-derived MSCs (AD-MSCs) and umbilical cord tissue-derived MSCs (UC-MSC) in a dextran sulfate sodium (DSS)-induced mouse model. Methods: Human AD-MSCs and UC-MSCs (1 × 106 cells) were injected intravenously into a 7-day DSS-induced colitis model. The therapeutic effects of cell origin, injection timing, and supernatants obtained from MSC cultures were evaluated. We also analyzed mRNA expression in MSCs, tissues, and intestinal flora. Results: AD-MSCs and UC-MSCs had strong anti-inflammatory effects when injected on day 3 in a mouse model. On day 11, mRNA levels of inflammatory factors in colon tissues were significantly decreased after injection of MSCs on day 3. Supernatants from MSCs culture decreased mRNA levels of tumor necrosis factor (Tnf)-α, but had reduced therapeutic effects compared with MSC cell injection. RNA sequencing using colon tissues obtained the day after cell injection revealed changes in the TNF-α/nuclear factor-κB and T-cell receptor signaling pathways. Additional analyses showed that several factors, including chromosome 10 open reading frame 54, stanniocalcin-1, and TNF receptor superfamily member 11b were increased in MSCs after adding serum from DSS colitis mice. Furthermore, both AD-MSCs and UC-MSCs maintained the balance of intestinal flora. Conclusion: AD-MSCs and UC-MSCs showed therapeutic effects against inflammation after early cell injection while maintaining the intestinal flora. Although supernatants showed therapeutic effects, cell injection was more effective against inflammation.

Differential gene expression following SCI in trkB.T1 null

Project description:We asked what genes are significantly differentially regulated in the spinal cord of SCI trkB.T1 WT and trkB.T1 KO mice. TrkB.T1 is upregulated shortly after SCI although the precise mechanisms underyling this upregulation are poorly understood. In the trkB.T1 null, we show less mechanical allodynia and better locomotor recovery following SCI. The microarray studies helped us to elucidate a signaling pathway that is differently regulated in the WT versus KO mice at 1 day after SCI. In this study, we did not examine gene changes within a genotype after SCI. Rather, we examined DGE by genotype at each time point. Spinal cord tissue from WT and KO mice in a sham condition (intact spinal cord) versus 1D, 3D and 7D following SCI was harvested for microarray analyses.

Single Cell Transcriptomic Analysis of Clinical-used MSCs Derived From Adipose Tissue, Bone Marrow, Placenta Membrane and Umbilical Cord

Project description:Here, we use single cell transcriptomics to characterize a dynamic senescence process of in vitro expanded MSC products from adipose tissue (AD), bone marrow (BM), placenta membrane (PM) and umbilical cord (UC). We found that cultured MSCs underwent progressive cell aging. As compared to perinatal MSCs (derived from PM and UC), adult MSCs (derived from AD and BM) showed higher degree of senescence and impaired immunosuppressive function. Moreover, we identified the transcriptional network regulating the intra-population functional diversity, which provides us guidance to drive reprogramming of determined MSCs lineage with specific cellular functions for a particular therapeutic intent.
