Microarray analysis of E14.5 embryonic bladder urothelium following conditional Tbx2/Tbx3 loss

ABSTRACT: The T-box transcription factors Tbx2 and Tbx3 play critical roles in tissue patterning and organogenesis, but their specific function in bladder epithelial development remains poorly understood. In this study, we investigated the transcriptional changes occurring in the embryonic bladder urothelium following the deletion or reduction of Tbx2 and/or Tbx3 in epithelial progenitors. We isolated GFP-positive urothelial cells from E14.5 embryos using fluorescence-activated cell sorting (FACS) from pooled mutant and control genotypes. Total RNA was analyzed by microarray hybridization to identify downstream target genes and molecular pathways regulated by Tbx2/3 during early bladder development.
