Interrogating DNA methylation associated with Lewy body pathology in a cross brain-region and multi-cohort study

ABSTRACT: In this study we investigated DNA methylation association with Lewy body pathology, profiled genome wide using this Illumina EPIC bead chip array in a set of brain samples sourced from the UK Brain Bank Network from centres at Newcastle, Imperial College London, University College London and Oxford. Data was generated from 779 samples, sourced from 406 unique donors (n = 387 Anterior Cingulate Cortex (ACC), n = 392 Prefrontal Cortex (PFC)). Following quality control, linear modelling approaches were used to assess DNA methylation loci association with Lewy body pathology as measured by the Braak staging criteria. This data was meta analysed alongside two prior established cohorts (Netherlands Brain Bank and Brains for Dementia Research).
