Endothelial Epac1 facilitates YAP/TAZ controlled melanoma growth and angiogenesis


ABSTRACT: Mice were treated by 75 mg/kg BW/day tamoxifen intraperitoneally for 5 days to induce endothelial specific Epac1/Rapgef3 knockout. This knockout allows us to study the role of endothelial Epac1 in tumor angiogenesis in mice inoculated subcutaneously with B16-F10 melanoma. Tumour endothelial cells were then isolated from primary subcutaneous melanoma by MACS plus FACS sorting and processed for RNA isolation with RNAeasy micro kit. RNA sequencing for low input sample was performed by DNBSEQ-G400 platform based on a Switching Mechanism at the 5′ end of RNA Template (SMART) method.
