Transcriptomic Profiling of Mitochondrial RNA in HepG2 Cells Exposed to GenX, a Short-Chain PFAS


ABSTRACT: This study investigated the transcriptomic effects of GenX on the mitochondria of human liver HepG2 cells. Cells were exposed to GenX at 10 µM and 100 µM concentrations for 48 hours. Mitochondria were isolated and mitochondrial RNA was extracted using TRIzol. High-quality RNA samples were sequenced using ribosomal-RNA-depleted libraries on the Illumina NovaSeq platform. Per and polyfluoroalkyl substances (PFAS) such as hexafluoropropylene oxide dimer acid (GenX) are emerging environmental contaminants with increasing concern regarding their potential mitochondrial toxicity. This BioProject investigated the effects of GenX exposure on mitochondrial gene expression in human hepatic HepG2 cells. Cells were treated with GenX at 10 µM and 100 µM concentrations for 48 hours, followed by mitochondrial isolation and RNA sequencing of mitochondrial transcripts. Mitochondrial transcriptomic analysis revealed regulation of genes involved in oxidative phosphorylation, redox homeostasis, and apoptotic signaling. This dataset provides a resource for understanding PFAS induced mitochondrial dysfunction and hepatotoxicity mechanisms.
