RNA sequencing upon CD146 knockdown/overexpression

ABSTRACT: RNA sequencing analysis was performed on liver cancer cells upon genetic manipulation of their CD146 levels. For these assays, total RNA samples were prepared using TRIzol reagent from Hep3B cells infected with lentiviruses expressing Tet-Control shRNA or Tet-CD146 shRNA, and HepG2 cells infected with lentiviruses expressing pLVX or CD146oe. The library and transcriptome sequencing were analyzed by AZENTA Life Sciences (Suzhou, China). Gene set enrichment analysis was applied to analyze the gene expression signatures using the Gene Ontology, Hallmark, and KEGG gene sets.

Project description:Metastasis is a complex process involving loss of adhesion, migration, invasion and proliferation of cancer cells. Cell adhesion molecules play a pivotal role in this phenomenon by regulating cell-cell and cell-matrix interactions. CD146 (MCAM) is associated with advanced tumor stage in melanoma, prostate and ovarian cancers. RNA-seq of CD146-positive vs CD146-negative cells isolated from human meniscus

Project description:This study aims to characterize the transcriptomic differences between CD146-positive and CD146-negative cells isolated from human meniscus tissues. Meniscus tissue samples were collected from total knee arthroplasty (TKA) patients lacking significant joint space narrowing. CD146-positive and CD146-negative cells were isolated from each patient's meniscus tissue, yielding a total of 6 samples (3 CD146-positive and 3 CD146-negative). Total RNA was extracted using Trizol reagent, and cDNA libraries were constructed using the VAHTS Universal V6 RNA-seq Library Prep Kit for Illumina following poly(A) mRNA selection. Libraries were sequenced on the DNBSEQ-T7 platform with 150 bp paired-end reads. Raw sequencing reads were quality-filtered and aligned to the human reference genome GRCh38 (Ensembl104) using HISAT2. Gene-level counts were obtained with HTSeq-count, and differentially expressed genes between CD146-positive and CD146-negative cells were identified using DESeq2 with thresholds of |fold change| > 2 and FDR < 0.05. Functional enrichment analyses including Gene Ontology (GO) and KEGG pathway analysis were performed to interpret the biological significance of the differentially expressed genes.

CD146 accelerates malignant progression of phyllodes tumor cells

Project description:Breast malignant phyllodes tumors (PTs) are rapid-progressing tumors, known to lack effective treatment and suitable prognostic markers as well as elaborate studies. Herein,we found that level of CD146 is progressively elevated with malignant progression of PTs and is an independent predictor for prognosis of PT patients.Mechanistically, CD146 activates the PI3K/AKT signaling pathway, thereby enhancing malignant behaviors of PT cells.This study unveils a CD146-PI3K/AKT axis in modulating the malignant progression of PTs, and open avenues to develop a novel target for precise prognosis and treatment for breast malignant PTs.

CD146 expression in mesenchymal stem cells is associated with vascular smooth muscle commitment

Project description:Bone-marrow mesenchymal stem cells (MSCs) are plastic adherent cells that can differentiate into various tissue lineages, including osteoblasts, adipocytes and chondrocytes. However, this progenitor property is not shared by all cells within the MSC population. In addition, MSCs vary in their proliferation capacities and expression of markers. Because of heterogeneity of CD146 expression in the MSC population, we compared CD146-/Low and CD146High cells under clonal and non-clonal (sorted MSCs) conditions to determine whether this expression is associated with specific functions. CD146-/Low and CD146High MSCs did not differ in colony-forming unit-fibroblast number, osteogenic and adipogenic differentiation or in vitro hematopoietic supportive activity. However, CD146-/Low clones proliferated slightly but significantly faster than did CD146High clones. In addition, a strong expression of CD146 molecule was associated with a commitment towards a vascular smooth muscle cell lineage with upregulation of calponin-1 expression. Thus, within a bone-marrow MSC population, certain subpopulations characterized by high expression of CD146, are committed toward a vascular smooth muscle cell lineage.

Comparison of periosteal, adipose, and dermal CD146+ human pericytes

Project description:Human pericytes demonstrate multilineage differentiation potential, and their descendants participate in tissue homeostasis and repair. Increasing evidence from developmental biology and tissue engineering suggest that regional specification by tissue of origin exists among human pericytes. Here, we sought to define the differentiation of CD146+ human pericytes from skeletal and soft tissue sources. Uncultured CD146+CD31-CD45- pericytes were derived by fluorescent activated cell sorting from human periosteum, adipose, or dermal tissue. Periosteal CD146+CD31-CD45- cells retained canonical features of pericytes, including cell surface marker expression, multilineage differentiation potential, and paracrine induced tubulogenesis. Periosteal pericytes demonstrated a striking tendency to undergo osteoblastogenesis, while soft tissue pericytes did not in vitro or in vivo. Microarray analysis demonstrated substantive differences between periosteal pericytes in comparison to their soft tissue pericyte counterparts. In sum, skeletal and soft tissue pericytes differ in their relative lineage differentiation potential and ability to form bone. Human tissues were microdissected, digested, and FACS sorted to derived a CD146+CD31-CD45- pericyte population, and cells were expanded in standard growth medium (DMEM, 10% FBS, 1% pen/strep) and nucleic acid isolated at subconfluency
