Macrophage TREM2 is a triglyceride-rich lipoprotein receptor that undergoes shedding in prediabetic hepatic steatosis [P630-6]


ABSTRACT: A mouse model of diet-induced prediabetes and hepatic steatosis was established using male LDL receptor-deficient (Ldlr-/-) mice fed a diabetogenic diet with added cholesterol (36% fat, 36.2% sucrose, 0.15% added cholesterol; DDC). Compared with littermate controls fed a standard laboratory diet (SLD), mice fed DDC for 22 weeks exhibited elevated plasma sTREM2, APOC3, triglycerides, cholesterol, and free fatty acids, as well as a positive correlation between plasma sTREM2 and APOC3. Liver bulk RNA sequencing showed increased macrophage and inflammatory signatures and reduced cholesterol biosynthesis signatures consistent with hepatic cholesterol accumulation in this mouse model of prediabetes and hepatic steatosis.
