Expression data form EhCPBF1 gene silenced strain.

ABSTRACT: Enteric protozoan parasite Entamoeba histolytica is a causative agent of amebiasis. One of the major virulence factors of this parasite is cysteine protease (CP). To understand the molecular basis of intracellular trafficking of CP, we identified CP5 binding protein and named cysteine protease binding protein family 1 (CPBF1). To study the function of CPBF1, we generate CPBF1 gene silenced strain and analyzed the expression profile by custom made microarray.

Project description:Enteric protozoan parasite Entamoeba histolytica is a causative agent of amebiasis. One of the major virulence factors of this parasite is cysteine protease (CP). To understand the molecular basis of intracellular trafficking of CP, we identified CP5 binding protein and named cysteine protease binding protein family 1 (CPBF1). To study the function of CPBF1, we generate CPBF1 gene silenced strain and analyzed the expression profile by custom made microarray. E. histolytica G3 strain was transfected with pSAP2 vector or pSAP2-CPBF1 and established the transformats by selecting the transfected cells with G418. After confirm the gene silencing of CPBF1 by RT-PCR, we performed the transcriptome analysis of pSAP2 (mock) and pSAP2-CPBF1 (gene silenced) cells.

Among them, CPBF2 gene silence caused defect in Matrigel invasion. To clarify transcriptomic difference in CPBF2 gene silenceing strain, RNA-seq analysis was conducted.

RNA-Seq expression analysis of Entamoeba histolytica trophozoites adapted to grow in the presence of toxic amount of the Nitric Oxide donor S-Nitrosoglutathione (GSNO)

Project description:Entamoeba histolytica is a protozoan parasite responsible for amebiasis, a disease which is characterized by acute inflammation of the colon. Adaptation of the parasite to toxic level of nitric oxide (NO) produced by phagocytes may be essential for the establishment of chronic amebiasis and for the survival of parasite within the host. To obtain insight into the mechanism of adaptation to NO, E. histolytica trophozoites were selected in vitro by stepwise exposures to increasing amounts of NO donor S-Nitrosoglutathione (GSNO) up to a concentration of 110 µM. These NO adapted trophozoites (NAT) were more resistance to acute exposure of GSNO (350μM), to activated macrophages and have a better capability to invade porcine colon explants compared to wild-type trophozoites. The transcriptome of NAT was investigated by RNA-sequencing (RNA-seq) and it showed only a weak overlapping with the transcriptome of trophozoites exposed to acute nitrosative stress (TEANS).

Phagosome proteome of mock and Atg8 silenced Entamoeba histolytica strains

Project description:The enteric protozoan parasite Entamoeba histolytica have high phagocytic ability. Phagocytosis is also important for the pathogenicity of this parasite; molecular mechanisms of phagocytosis and phagosome maturation is focused. Atg8 is well studied autophagy marker protein. We previously shown that E. histolytica Atg8 translocate to nascent trogosomes and expression silencing of the Atg8 caused retardation of phagosome acidification. To investigate how Atg8 regulates phagosome maturation, here we conducted proteome analysis of phagosomes isolated from an E. histolytica strain in which atg8 gene expression are silenced (atg8 gene silenced, atg8-gs) and its mock control strain (transfected with psAP2-Gunma).

Project description:Entamoeba histolytica is a protozoan parasite which causes colitis and liver abscesses. Using a genomic DNA microarray consisting of 1.6 - 2.0 kb genomic inserts we have generated a transcriptional profile of 1,971 unique parasite transcripts. The arrays in this experiment set were used to (1) estimate relative transcript abundance for Entamoeba histolytica (HM-1:IMSS) trophozoites in mid-logarithmic growth and (2) to examine changes in the transcriptional profile of Entamoeba histolytica when the parasite interacts with colonic epithelial cells (Caco-2). A time course was used such that RNA was isolated from ameba alone and ameba + Caco-2 cells at 3hrs, 6hrs and 9hrs corresponding to 10%, 50% and 90% cell monolayer destruction. At least two biological experiments and three replicates were used for each time point. Groups of assays that are related as part of a time series. Keywords: time_series_design

RNA-seq analysis of Entamoeba histolytica glycerol 3-phosphate dehydrogenase (G3PDH) and glycerol kinase (EhGK) gene silencing strain

Project description:Amebiasis is caused by the protozoan parasite Entamoeba histolytica. Treatment options other than metronidazole and its derivatives are few, and their low efficacy against asymptomatic cyst carriers, and experimental evidence of resistance in vitro justify the discovery/repurposing campaign for new drugs against amebiasis. To better understand the role of glycerol metabolism in this parasite, we focused on characterizing two important enzymes, glycerol kinase (GK) and glycerol 3-phosphate dehydrogenase (G3PDH). Recombinant GK was biochemically characterized in detail, while G3PDH was not due to failure of protein expression and purification. GK revealed novel characteristics and unprecedented kinetic properties in reverse reaction. Gene silencing revealed that GK is essential for optimum growth, whereas G3PDH is not. Gene silencing of G3PDH caused upregulated GK expression, while that of GK resulted in upregulation of antioxidant enzymes as shown by RNA-seq analysis. Although the precise molecular link between GK and the upregulation of antioxidant enzymes was not demonstrated, the observed increase in antioxidant enzyme expression upon GK gene silencing suggests a potential connection between GK and the cellular response to oxidative stress. Together, these results provide the first direct evidence of the biological importance and coordinated regulation of the glycerol metabolic pathways for proliferation and antioxidative defense in E. histolytica, justifying the exploitation of these enzymes as future drug targets.
