ScRNA-seq of adipose tissue from insulinoma patients and healthy controls


ABSTRACT: We performed single-cell RNA sequencing of paired subcutaneous and visceral adipose tissue from patients with insulinoma and healthy controls to investigate insulin-dependent mechanisms of adipose-tissue cell differentiation in vivo. Insulinoma provides a unique clinical model of chronic endogenous hyperinsulinemia, enabling the assessment of cellular and transcriptional responses to sustained insulin exposure in human adipose tissue. Stromal vascular fraction cells were profiled to characterize tissue-resident cell populations and to identify insulin-associated changes in cell-state composition, differentiation trajectories, and gene-expression programs. By comparing subcutaneous and visceral depots, we further examined whether hyperinsulinemia-associated mechanisms differ between anatomically and metabolically distinct adipose tissues. This study provides a single-cell resource for investigating depot-specific regulation of adipose tissue remodeling and may help clarify the role of insulin signaling in human adipose cell differentiation.
