Comparative transcriptomic analysis of GPC3 wild-type and knock-out HepG2 and Hep3B hepatocellular carcinoma cells 6 hours post-radiation

ABSTRACT: Glypican-3 (GPC3) is a cell-surface oncofetal antigen heavily implicated in hepatocellular carcinoma (HCC) progression and resistance to therapy. To elucidate the role of GPC3 in modulating radiation response, total RNA sequencing was performed on GPC3 wild-type (WT) and GPC3 knock-out (KO) HepG2 and Hep3B cell lines harvested 6 hours after radiation exposure. Comparative transcriptomic profiling across these 4 irradiated hepatocellular carcinoma cell line models identifies differential gene expression programs and potential signaling pathways regulated by GPC3 upon ionizing radiation.
