Early transcriptional responses of mouse bone marrow-derived macrophages to Mycobacterium wolinskyi QL01

ABSTRACT: Mycobacterium wolinskyi is an emerging rapidly growing nontuberculous mycobacterium, but the early host response to this species remains poorly characterized. To investigate the macrophage response to M. wolinskyi, primary bone marrow-derived macrophages from C57BL/6J mice were infected with the clinical isolate QL01; matched uninfected macrophages served as controls. Poly(A)-selected RNA sequencing was performed with three biological replicates per group. Transcriptomic analysis revealed extensive differential gene expression and positive enrichment of TNF, NF-kappa B, C-type lectin receptor, and Toll-like receptor signaling pathways in QL01-infected macrophages. These data characterize the early innate immune and pro-inflammatory transcriptional response of macrophages to M. wolinskyi QL01.
