Transcriptomic Classification and Developmental Mapping of Canine Acute Leukemia Reveal Distinct Myeloid and Lymphoid Subgroups

ABSTRACT: Canine acute leukemias are biologically heterogeneous hematopoietic malignancies whose molecular features remain incompletely defined. We performed bulk RNA sequencing of peripheral blood leukocytes from 110 dogs with acute leukemia together with 10 immunophenotypically normal peripheral blood controls and 5 immunophenotypically normal bone marrow controls. The study was designed to define transcriptional programs associated with canine acute leukemia, resolve myeloid- and lymphoid-associated molecular subgroups, compare leukemia profiles with normal hematopoietic reference populations, and evaluate conserved developmental and leukemogenic programs relevant to comparative leukemia biology.
