Effect of finerenone treatment on the transcriptomic profile of conditionally immortalized human podocytes

ABSTRACT: Finerenone is a non-steroidal mineralocorticoid receptor antagonist that has demonstrated renoprotective effects in patients with chronic kidney disease, yet its actions in podocytes remain incompletely defined. To systematically explore the effects of finerenone on podocytes, we performed RNA sequencing of conditionally immortalized human podocyte (HPC) cells treated with 4 µM finerenone for 24 hours or vehicle, with three biological replicates per group. Transcriptomic profiling identified 716 differentially expressed genes (|log2FC| ≥ 1.5, P < 0.05), including 360 upregulated and 356 downregulated genes. Functional enrichment analyses indicated that these genes were involved in cytoskeletal dynamics, cell death, immune-inflammatory responses, and metabolic homeostasis, among other biological processes. Together, these data indicate that finerenone regulates podocyte gene expression through multiple pathways, providing transcriptomic evidence supporting its protective effects on podocytes.
