Single-cell RNA sequencing Deciphers the Immune Microenvironment Landscape of Ascites in Ovarian Cancer Mice Following Chemotherapy

ABSTRACT: To investigate the remodeling effects of chemotherapy on the immune microenvironment and its impact on ovarian cancer progression, we established a murine ovarian cancer chemotherapy model and performed 10x Genomics single-cell sequencing to systematically characterize the dynamic landscape of the ascites immune microenvironment before and after treatment. Although chemotherapeutic agents exert direct cytotoxic effects on tumor cells, their immunomodulatory actions may paradoxically promote tumor progression through microenvironmental remodeling. Previous sequencing studies using clinical samples have suggested that chemotherapy can induce the formation of an immunosuppressive microenvironment; however, these findings lack in vivo validation and mechanistic exploration in mouse models. This study provides, for the first time, a comprehensive single-cell resolution atlas of the ascites immune microenvironment in ovarian cancer mice before and after chemotherapy, offering novel insights and potential therapeutic targets for improving the prognosis of ovarian cancer patients.
