Single-cell RNA sequencing of Chlamydomonas reinhardtii reveals latent and lytic transcriptional programs of an integrated giant virus (GEVE)

ABSTRACT: Giant viruses of the phylum Nucleocytoviricota can integrate into the nuclear genomes of their algal hosts as giant endogenous viral elements (GEVEs). We generated single-cell RNA sequencing data (Parse Biosciences Evercode split-pool combinatorial barcoding) from Chlamydomonas reinhardtii strain CC-2937, which carries a ~618 kb integrated GEVE (punuivirus) on chromosome 15 encoding ~506 annotated genes. Samples were collected at days 4 through 7 post-inoculation (DPI4-DPI7), profiling 22,421 cells across two biological replicates per timepoint, to characterize the transcriptional dynamics of GEVE gene expression at single-cell resolution.
