ATACseq of ENTPD1/CD39 and NT5E/CD73 in A549 human NSCLC cell lines

ABSTRACT: There is a highlighted need to understand properties of the CD39/CD73 pathway in human lung malignancies considering recent clinical trials demonstrating prominent anti-tumor activity of CD73-blocking antibodies in combination with PD-L1 antagonists. To establish if proinflammatory cytokines commonly upregulated in the inflamed TME could modulate CD39 and CD73 expression levels, we exposed cultured human A549 lung ADC cells to IFNγ and TNFα and found an induction of CD73 protein expression. Using ATAC-seq, we analyzed DNA extracted from these control and treated cell preparations, with the goal to explore mechanisms underlying the increased CD73 expression in proinflammatory states.
