Ovarian RNA sequencing of female ICR mice following one month of 6PPDQ exposure

ABSTRACT: This study profiled ovarian gene expression in female ICR mice following oral exposure to 6PPDQ. Treatment began at 5 weeks of age. Six mice received 6PPDQ at 4 mg/kg by daily oral gavage for one month, and six control mice received corn oil by daily oral gavage for one month. Ovarian RNA-seq was used to compare gene expression between the treatment and vehicle-control groups.

Bisphenol A-induced renal metabolic disruption: Insights into MAPK-mediated steroid hormone dysregulation from transcriptomic and metabolomic analyses

Project description:After long-term exposure of BPA, human is probably suffering from invisible renal injury and metabolic disruption, while its linked intricate gene transcriptional mechanisms warrant more conclusive evidence. Comprehensive analysis utilizing transcriptomics and metabolomics were used to elucidate the transcriptional mechanisms regulating metabolic homeostasis involved in BPA-induced nephrotoxicity. Here, six-week-old male ICR (CD1) mice were randomly divided into vehicle group with 10 mice and BPA exposure group with 10 mice (five mice per cage). The mice in the exposure group received a daily gavage of BPA dissolved in an aqueous solution containing 0.1% ethanol at the dose of 50 μg/kg/d, which is the tolerable daily intake (TDI) dose of BPA. The mice in the vehicle group received the same dose of aqueous solution containing 0.1% ethanol. After 6-week exposure, the mice were euthanized and left kidney were tissues were frozen with liquid nitrogen and used for RNA transcriptome sequencing.

Investigation of gene expression alterations of pancreatic cancer caused by Deferasirox administration

Project description:To investigate the genetic effect of Deferasirox in pancreatic cancer, we examined gene expression alternations in the removed tumors exposed to Deferasirox. Pancreatic cancer cells (BxPC-3) in culture were harvested and viable cells were injected subcutaneously into the backs of the mice. When tumor volumes reached 150 mm3, treatment began. The treatment group of mice received Deferasirox suspended in saline, which was administered by oral gavage every second day, with three treatments per week, over 21 days at concentrations of 200 mg/kg. The control mice were treated with the vehicle alone. At the end of the experiment, the mice were sacrificed and the tumors were excised and processed for genetic analyses. A total of 6 tumors were analyzed. Of these, 3 tumors were removed from vehicle-treated mice, and the other 3 tumors were removed from Deferasirox 200 mg/kg-treated mice.

Effects of low-doses of antiandrogenic substances on Wistar Rat testes [Transcriptome analysis]

Project description:Three antiandrogenic substances (Flutamide, Prochloraz, Vinclozolin) were administered in aqueous preparations at 3 doses: i) Acceptable Daily Intake (ADI) ; ii) No Observed Adverse Effect Level (NOAEL) ; iii) Low Observed Adverse Effect Level (LOAEL) . Pregnant females (20/group) were treated by gavage from gestation day (GD) 6 until postnatal (PND) day 21. After weaning, the offspring received the same test-substance as their mothers by oral gavage. Twenty males were selected to be raised until the weaning (Subset 1, PND 21), the day of sexual maturation (Subset 2, puberty) or early adulthood (Subset 3, PND 83±2). At each subset, testes were used for transcriptome and miRNome analyses.
