Divergent Transcriptional Programs of EGF, Tremella polysaccharide, Aloe vera, and Dragon fruit extract Converge on Fibroblast Activation for Canine Wound Healing


ABSTRACT: Like humans, canine skin wound healing requires rapid fibroblast recruitment and activation. Here, we investigated the effects of whey epidermal growth factor (EGF), Tremella polysaccharide, aloe vera, and dragon fruit extract on wound healing in canine skin fibroblasts. All four agents stimulated proliferation and migration in vitro. RNA-seq of EGF- and Aloe-treated cells revealed distinct transcriptional programs: EGF up-regulated ribosomal structural genes and signaling-receptor regulators, indicating enhanced protein synthesis and signal transduction, whereas Aloe preferentially induced genes involved in amide and peptide binding, implicating extracellular-matrix modulation. qRT-PCR confirmed that each extract elevated key wound-healing genes (CXCR1, FOXD1, FGF1, and COL8A2). Although these ingredients have different chemical properties, EGF, Tremella polysaccharide, Aloe vera, and Dragon fruit extract can all promote fibroblast proliferation and migration, providing a theoretical basis for promoting wound healing in canine skin.
