Colon T cells during T muris and C rodentium infections [I28]

ABSTRACT: This study is part of the immgenT Open Source Project, specifically from IGT28, which focuses on characterizing colon T cells during Trichuris muris or Citrobacter rodentium infections in mice. 9 weeks old male and female mice were infected via oral gavage with T. muris (20 eggs, analyzed at 35 days post-infection) or C. rodentium (10^8 CFU, analyzed at 14 days post-infection).

Project description:We report that dietary iron protects mice from infection by Citrobacter rodentium. Iron induces a state of insulin resistance and increases glucose availability in the gut, thereby attenutating C. rodentium virulence. Additionally the pathogen appears to be driven towards a long-term commensal state. Here, we identify mutations in persistent and avirulent Citrobacter rodentium isolates from mice given an iron supplemented diet.

Project description:Trichuris muris, the mouse whipworm is used as a laboratory model of the human parasite T. trichuira - a gastrointestinal nematode. Three laboratory isolates of T. muris exist: the E, J and S isolates. Previous data has shown that the S isolate survives to chronicity in C57BL/6 mice unlike the E and J isolates which are expelled. The ability of the S isolate to persist is still unclear. Microarray analysis of gut samples from mice infected with either the E or S isolates of T. muris for 7 days was used to determine early changes in gene expression that may later determine the outcome of infection.
