Project description:This study is part of the immgenT Open Source Project, specifically from IGT40, which focuses on a pan-organ and time course analysis of T cells in acute LCMV (Armstrong) infection at day 60 post-infection. The dataset includes T cells isolated from multiple organs: spleen, prostate, submandibular gland, bone marrow, mesenteric lymph node, small intestine epithelium, small intestine lamina propria, lung, and mediastinal lymph node. 7-week-old male C57BL/6J mice infected intraperitoneally with 2×10^5 PFU LCMV Armstrong, and T cells were analyzed at 60 days post-infection (dpi). The study includes both endogenous T cells and adoptively transferred P14 and SMARTA TCR transgenic T cells. Mice received IV 100,000 SMARTA (CD45.1.2) and 50,000 P14 (CD45.1) T cells one day before infection.

Genome-wide profiling of early and late CD8 memory T cells expressing CD62L

Project description:Memory CD8+ P14 cells were generarted through adoptive transfer and infection with LCMV armstrong. Then, early memory (after 30 - 45 days) and late memory (after 8 months) cells were sort purified based on CD62L expression. Genome-wide expression profiles were captured for early and late memory cells with high and low levels on CD62L using Affymetrics arrays to show their molecular and functional differences.

Project description:Lymphocytic Choriomeningitis Virus (LCMV) specific CD8+ T cells (P14) were transferred into congenic WT mice followed by LCMV(DOCILE) infection. CXCR5-expressing (CXCR5+) or CXCR5 non-expressing (CXCR5-) P14 were purified on day 8 after infection, and total mRNA were sequenced from these populations. mRNA of P14 from uninfected mice (Naive P14) was also sequenced. Examination of mRNA level in CXCR5 expressing P14 (CXCR5+P14) and non-expressing P14 (CXCR5-P14) from LCMV infected mice day 8 post infection. mRNA of P14 from uninfected mice (NaÃ¯ve P14) was also examined.

RNAseq analysis of PS+ vs. PS- effector CD8 T cells sorted from spleen of LCMV-Armstrong infected mice

Project description:We report the transcriptional profile of phosphatidylserine (PS) positive and PS negative CD44+CD62L- effector CD8+ T cells from LCMV-Armstrong infected mice sorted from spleens on day5 amd day10 after infection. PS was stained in vivo by injection of MFG-E8-eGFP. PS+ cells acquire PS through binding of extracellular vesicles (EVs). Binding of EVs by effector CD8+ T cells is strongly enhanced during LCMV infection and peaks between day5 and day10 post infection. Effector T cell gene signature is increased in EV+ CD8+ T cells compared to their EV- counterparts.

RNA sequencing of wild type and FXR KO CD8+T cells in fed/fasted LCMV-infected mice

Project description:Conditional ablation of FXR in T cells was achieved with CD4-Cre Nr1h4fl/fl mice. Twenty thousend congenically marked naive (CD44- CD62L+) WT and FXR KO T cells carrying the OT-I transgenic TCR were cotransferred into lymphoreplete hosts. Recipients were challenged with LCMV-OVA and allowed to feed Ad libitum (AL) or fasted (Fs) for 24h beginning at 6pm on D5 post-infection. Splenic CD44+ effector cells of each genotype were FACS-purified (double-sorted) at the end of the starvation period.
