Comparison of editing efficiencies of intein, AAVLINK, and StitchR delivery ABE8e systems at the mouse TA

ABSTRACT: In this study, intein, AAVLINK, and StitchR delivery ABE8e systems were packaged into viral vectors (AAV) and intramuscular injected into mice. Genomic DNA was extracted from the mouse tibialis anterior (TA) muscle, followed by deep sequencing to analyze the editing efficiency.

Transcriptomic analysis of PAX7+ cells isolated from injured Tibialis Anterior muscles of mice exposed to hypoxic versus normoxic atmosphere

Project description:Acute muscle injury was induced by cardiotoxin intramuscular injection in the tibialis anterior (TA) of 8-week-old Tg:Pax7nGFP mice. Mice were then housed in standard normoxic atmosphere (21%O2) or in a normobaric hypoxic chamber (10%O2), from 0 to 7 days post-injury (dpi). FACS-sorted PAX7+ (GFP+) cells were isolated from CTX-injured TAs at 7 dpi to perform a bulk RNAseq analysis.

Project description:To investigate the role of the circadian clock gene Bmal1 in skeletal muscle, we compared the circadian transcriptomes of fast tibialis anterior (TA) and slow soleus (SOL) skeletal muscles from muscle-specific Bmal1 KO (mKO) and their control Cre- littermates (Ctrl). Keyword: Circadian Transcriptome, time course 72 samples were analyzed, comprised of 4 experimental groups (Ctrl SOL, mKO SOL, Ctrl TA, mKO TA), with 3 biological replicates for each time point sampled every 4 hours for 24 hours. Project description:To evaluate decellularized skeletal muscle extracellular matrix hydrogel effectiveness in treating disuse atrophy using a mouse hindlimb suspension (HU) model. Project description:We applied a 5'RNA-seq methodology to assess gene and differential isoform expression in striated muscle tissues extracted from adult wild-type mice. 5'RNA-seq analysis of transcriptomes from mouse soleus, tibialis anterior (TA), diaphragm and left ventricle myocardial tissues. Three biological replicates per tissue were pooled into a single sequencing run. 5'RNA-seq methodology consists of enhanced sequencing of 5' ends and computational assessment of changes at start-sites of genes.

Profiling of glycerol-induced models of muscle regeneration upon melatonin treatment for 14 days in mice

Project description:Intramuscular injection of glycerol (Gly) has been reported to lead to skeletal muscle regeneration and ectopic fat deposition. We utilized glycerol injections in the tibialis anterior muscles to investigate the effects of melatonin on skeletal muscle regeneration and intramuscular fat infiltration. We characterized genome-wide expression profiles of tibialis anterior muscles from wild-type mice injured by glycerol injection following melatonin intervention. Mice that were treated with melatonin exhibited improved muscle regeneration and reduced intramuscular fat deposition, which were associated with enhanced myogenesis, remodeled lipid metabolism and reduced immune cell infiltration.
