Immune-related gene expression signatures associated with progression of myelodysplastic neoplasms

ABSTRACT: Myelodysplastic neoplasms (MDS) are clonal hematopoietic stem cell disorders with wide prognostic variability. Here, we aimed to identify gene expression patterns associated with MDS progression and to characterize the underlying biological pathways. We conducted a retrospective case–control study of 71 patients initially diagnosed with lower-risk MDS: 31 subsequently developed MDS with increased blasts (MDS-IB) or acute myeloid leukemia (AML), whereas 40 remained stable. Archival formalin-fixed, paraffin-embedded (FFPE) bone marrow samples were profiled using a NanoString panel of 825 genes related to oncogenesis, immune response, and leukemic transformation. We compared diagnostic samples from patients with progressive vs. stable disease, as well as paired diagnostic and progression samples.
