Project description:The aim of this study was to quantify the impact of NOD genetic vatiation on thymic negative selection transcriptional programs. Pre-selected BDC2.5 TCR Tg DP thymocytes from non-selecting B6 and NOD.H2b backgrounds were purified (Dynal CD8 FlowComp), mixed in a 1:1 ratio and stimulated with BDC mimotope-loaded TCRa-/-/NOD splenocytes for indicated periods of time and double sorted by FACS as Thy1.2+Dump-CD4+CD8+; Dump includes CD19, Gr1, CD11b, CD11c, CD49b. Profiling of aggressive vs benign pancreatic infiltrates in the BDC2.5 Tg model of Type I diabetes

Project description:This study was performed to understand what controls the aggressivity of the pancreatic infiltrate during type-I diabetes development. We used the BDC2.5 transgenic mouse model. Samples were obtained at the age of onset of insultis. Depending on their genetic background, mice transgenic for the BDC2.5 T cell receptor present very different forms of insulitis. The NOD genetic background leads to a benign insulitis whereas the C57Bl/6-H2g7/g7 leads to an aggressive insulitis. We first studied how antigen-specific T cells are affected by these differences by obtaining the transcriptional profiles of BDC2.5 T cells from pancreas and pancreatic lymph nodes. We also compared the gene expression profiles of the entire leukocyte population present in the pancreatic lesion. Keywords: other

Transcription profiling time series of thymocytes from HNT-TCR transgenic mice injected with HA peptide to induce thymic negative selection

Project description:During T cell development in the thymus, negative selection eliminates autoreactive and potentially harmful T cells, leading to central tolerance. This process, involves specific apoptosis programs thought to be governed by the transcription factor Nur77. To analyze the genetic pathways involved in thymic negative selection and to point to new genes involved in thymic selection, we performed a high throughput temporal expression profiling of thymocytes from HNT-TCR transgenic mice undergoing specific peptide-mediated thymic negative selection, using oligonucleotide microarrays. We used HNT-TCR transgenic mice that are on a B10D2 background and express a transgenic TCR (V beta 8), specific for the HA peptide (HNTNGVTAACSHE) presented by MHC class II I-A^d. In this model of thymic negative selection, a single injection of the specific HA peptide induces massive and synchronized deletion of thymocytes. In this study, we analyzed the dynamically regulated genes over a 24 hours period, with a focus on the early time points of 3 and 6 hours after peptide injection.
