Genome-wide analysis of mouse macrophages stimulated with IL-4 (bone marrow macrophages) (Affymetrix)

ABSTRACT: Analysis of alternative activation of macrophages at gene expression level. The study forms part of a wider study where we compare the effects of IL-4 in different human and mouse macrophages. Our results support the notion that in vitro culture conditions greatly affect the macrophage response to IL-4. Total RNA obtained from bone marrow derived macrophages upon exposure to 20 ng/ml of IL-4 for 18 hours.

The immunomodulatory effects of IL-4 on monocyte-derived macrophages

Project description:To explore the effects of IL-4 on tumor-associated macrophagess, we first sorted CD14+ monocytes from peripheral blood mononuclear cells of healthy donors using magnetic-activated cell sorting and then induced their differentiation into macrophages by culture with M-CSF for 5 days. We then stimulated these macrophages with or without IL-4 (20 ng/ml) for 6 hours and examined gene expression using a cDNA microarray. IL-4 stimulated gene expression on monocyte-derived macrophages was measured at 6 hours after exposure to doses of 20 ng/ml IL-4.

High salt reduces the activation of IL-4+IL-13 stimulated macrophages

Project description:High salt (NaCl) intake is a major risk factor for cardiovascular disease; high NaCl has also been shown to be a powerful modulator of pro-inflammatory immune cell polarization. To investigate the effect of high NaCl on the polarization of M(IL-4+IL-13) macrophages we performed gene expression analysis by microarray and genome wide histone modification (H3K4me3 and H4ac) analysis by ChIP-seq of murine bone marrow derived macrophages. Macrophages were polarized with recombinant IL-4 and IL-13 in vitro in the presence or absence of an additional 40mM of NaCl in the culture medium for 24h before analysis.
