Array CGH from mouse SFME cells and SFME cells differentiated with TGFB and FCS

ABSTRACT: SFME cells (ATCC) were grown as spheres or differentiated with TGFß for 24 h or 10%FCS for 12 h. ArrayCGH experiments were performed on all three samples.

Fucose-mediated transcriptional activation of the fcs operon by FcsR in Streptococcus pneumoniae

Project description:In this study, we explore the impact of fucose on the transcriptome of S. pneumoniae D39. The expression of various genes and operons, including the fucose uptake PTS and utilization operon (fcs operon), was altered in the presence of fucose. By means of quantitative RT-PCR and β-galactosidase analysis, we demonstrate the role of the transcriptional regulator FcsR, present upstream of the fcs operon, as a transcriptional activator of the fcs operon. We also predict a 19-bp putative FcsR regulatory site (5’-ATTTGAACATTATTCAAGT-3’) in the promoter region of the fcs operon. The functionality of this predicted FcsR regulatory site was further confirmed by promoter-truncation experiments, where deletion of half of the FscR regulatory site or full deletion led to the abolition of expression of the fcs operon.

Expression data in patients with familial (FCS) and multifactorial chylomicronemia (MCM)

Project description:FCS and MCM appear highly similar disorders. However, they importantly differ in term of risk and response to treatments. Some sets of genes might be up or down regulated in both disorders and could partly explain those differences. We used microarrays to explore gene expression patterns in FCS and MCM patients to identify potential targets that can be selected for protein quantification and functional studies.

Project description:arrayCGH profiling of 10 cell lines derived from clear cell Renal Cell Carcinoma The goal of this study was to make a detailed genomic profile of these cell lines, including aCGH, miRNA expression, and mRNA expression profiles. The miRNA and mRNA expressiion profiles are submitted to GEO separately. One paper is in preparation to link the miRNA expression data with the arayCGH data. Implication of Nos2 inactivation on genomic changes in Ptch1+/- medulloblastomas (array-CGH)

Project description:The Ptch1+/- strain constitues an established mouse model for the Shh-driven type of medulloblastoma. Combined Ptch1+/- Nos2-/- mice show a two-fold increased incidence for this tumor. Here, the impact of Nos2 inactivation on copy number alterations during medulloblastoma development was investigated by array-based comparative genomic hybridization (arrayCGH) of tumor samples from both genotypes.
