Primary B-lymphocytes were purified from mouse spleens and stimulated with IL-4 (Pepro-Tech) and CD40L (R&D Systems) as indicated at 5 ng/ml and 200 ng/ml, respectively. The BCR of stimulated cells was activated by incubation with goat F(abM-bM-^@M-^Y)2 anti-mouse IgM (Southern Biotechnologies) at 2.5 M-NM-<g/ml for 3 hour.

Project description:To elucidate the genome-wide role of Ca2+/ calmodulin inhibition of E2A in regulation of BCR activation, we performed DNA microarray analysis of activated splenic B cells expressing wildtype or CaM-resistant E12 mutant m8N47 with and without anti-IgM treatment for 3 hour.The expression of a remarkably large set of genes differed significantly. Primary splenic B-lymphocytes were purified from mice heterozygous for the deletion of E2A gene and infected with retroviruses expressing either wildtype or mutant E12 after activation with CD40L plus IL-4 for 14 h. After 12 h incubation, the infection was repeated for 12 h, followed by incubation for a further 22 h post-infection in fresh complete medium with the stimulants to allow expression of GFP and E12. In addition to CD40L plus IL-4, the medium was supplemented with LPS (200 ng/ml) during retroviral infection incubations to improve infection efficiency. To study gene expression following B-cell receptor activation, anti-mouse IgM (2.5 M-NM-<g/ml) was added for 3 hour. For DNA microarray analysis of infected B-lymphocytes, the infected cells were purified with a FACSAria cell sorter instrument (BD Biosciences) using their green fluorescence from expression of the GFP.

NFATc1 controls the survival, function and suppressive capacity of B lymphocytes upon B cell receptor stimulation

Project description:Triggering of B cell receptors (BCR) induces a massive synthesis of NFATc1 in splenic B cells. By inactivating the Nfatc1 gene and re-expressing NFATc1 we show that NFATc1 levels are critical for the survival of splenic B cells upon BCR stimulation. NFATc1 ablation led to decreased BCR-induced Ca++ flux and proliferation of splenic B cells, increased apoptosis and suppressed germinal centre formation and immunoglobulin class switch by T cell-independent antigens. By controlling IL-10 synthesis in B cells, NFATc1 supported the proliferation and IL-2 synthesis of T cells in vitro and appeared to contribute to the mild clinical course of Experimental Autoimmune Encephalomyelitis in mice bearing NFATc1-/- B cells. These data indicate NFATc1 as a key factor controlling B cell function. Splenic mice cells were isolated from mice bearing NFATc1 deficient B-cells and from control mice, stimulated with anti-IgM for 0h, 3h, 8h and 16h, respectively and isolated using Milteny beads to enrich the B cell population. This experiment was performed in 3 biological replicates.

Project description:To simulate transient B cell activation that is the likely initiator of T-dependent responses, we examined the molecular and functional consequences of a single-round of immunoglobulin M (IgM) signaling. This form of activation triggered early cytosolic signaling and transcription factor NF-kB activation indistinguishably from conventional continuous IgM cross-linking, but did not induce G1 progression. However, single-round IgM signaling changed the expression of chemokine and chemokine receptor genes implicated in initiating T-dependent responses, as well as accentuated responsiveness to CD40 signaling. Several features of single-round IgM signaling in vitro were recapitulated in B cells after short-term exposure to antigen in vivo. We propose that transient BCR signals prime B cells to receive T cell help by increasing the probability of B-T encounter and creating a cellular environment that is hyper-responsive to CD40 signaling. Primary B lymphocytes were isolated using Auto-MACS (Miltenyi Biotec) by negative selection. B cell purity was 90-95% based on flow cytometric analysis with CD19 staining. Purified B cells (2x10^6/ml) were cultured in RPMI 1640 supplemented with 10% heat-inactivated FBS, 55nM beta-mercaptoethanol, 2mM L-glutamine and 100IU penicillin and 100ug/ml streptomycin at 37degrees C. For pulsed anti-IgM treatment experiments, B cells were incubated with 10ug/ml goat anti-mouse IgM F(abâ)2 (Jackson ImmunoResearch Laboratories) at 4 degrees C for 30 min. Unbound anti-IgM was removed from the medium by washing and centrifuging the cells at 4 degrees C. The cells were resuspended in chilled complete medium and shifted to 37 degrees C by placing in an incubator or in water-bath. For continuous anti-IgM treatment experiments, B cells were stimulated with 10ug/ml anti-IgM at 4 degrees C for 30 min, then incubated at 37 degrees C.
