ABSTRACT: Transgenic GFAP-, NES-, and SYN- CRE mice were injected with a lenti-viral construct containing a floxed RFP directly upstream of a cassette containing si-p53, GFP, and mutant HRAS. Tumors arising from the various CRE tissue specific promoters and differing injections sites were compared to normal hippocampus and cortex.

Flow activated cell sorting and genome-wide analysis of hybrids cells (RFP+YFP+) and non-hybrid cells (RFP+)

Project description:Cellular functions in mouse and human tissues can be restored after fusion of bone marrow (BM)-derived cells with a variety of somatic cells. Here, after transplantation of hematopoietic stem and progenitor cells (HSPCs) in the substantia nigra pars compacta (SNpc) of two different mouse models of Parkinson’s disease, we significantly ameliorated the dopaminergic neuron loss and function. We show fusion of transplanted HSPCs with neurons and with glial cells in the ventral midbrain of Parkinson’s disease mice. Interestingly, the hybrids can undergo reprogramming in vivo and survived up to 4 weeks after transplantation, while acquiring features of mature astroglia. These newly generated astroglia produced Wnt1 and were essential for functional rescue of the dopaminergic neurons. Our data suggest that glial-derived hybrids produced upon fusion of transplanted HSPCs in the SNpc can rescue the Parkinson’s disease phenotype via a niche-mediated effect, and can be exploited as an efficient cell-therapy approach. To further investigate the reprogrammed phenotype of the hybrids genome-wide, we analyzed the transcriptome of the RFP+YFP+ fused cells compared to the RFP+ unfused cells using microarrays in the R26Y, GFAP-Cre and FoxA2-Cre models

RNA-seq of human midbrain organoid models of Parkinson's Disease

Project description:Parkinson’s disease (PD) is a prevalent neurodegenerative disorder that is characterized by the selective loss of midbrain dopamine (DA)-producing neurons and the formation of α-synuclein (α-syn)-containing inclusions named Lewy bodies (LBs). Here, we report that the loss of glucocerebrosidase (GCase), coupled with α-syn overexpression, result in substantial accumulation of detergent-resistant α-syn aggregates and Lewy body-like inclusions (LBLIs) in human midbrain-like organoids (hMLOs). These LBLIs exhibit a highly similar structure to PD-associated LBs, by displaying a spherically symmetric morphology with an eosinophilic core, and containing α-syn and ubiquitin. Importantly, hMLOs generated from PD patient-derived inducible pluripotent stem cells (iPSCs) harboring SNCA triplication also exhibit subsequent degeneration of DA neurons and LBLI formation upon chronic GCase inhibitor treatment. Taken together, our hMLOs harbouring two major PD risk factors (GCase deficiency and overproduced α-syn) successfully recapitulate major pathophysiological signatures of the disease, and highlight the broad utility of brain organoid technology in modeling human neurodegenerative diseases.

Project description:CMs were isolated from Myh6-Cre/Esr1;R26-tdTomato mice at P1 and P7,which were permanently labeled with RFP upon treatment with tamoxifen immediately after birth, and injected resulting CMs into infarcted mouse hearts induced by LAD ligation. After two weeks, RFP+ cells were isolated by FACS and subjected to scRNA-Seq.
