2 doses of ionising radiations (x-rays) -Transcriptomic analysis of effects of ionising radiation on Arabidopsis thaliana.

ABSTRACT: au09-03_gamma-irradiation - 2 doses of ionising radiations (x-rays) - Metabolic pathways involved in the response of plants to ionising radiation treatment. - Arabidopsis thaliana (Col-0) seeds were grown in Petri dishes under sterile conditions until they had 2-rosette-leaves (on average 10 days). Then they received a dose of 10 or 40 gray of X-ray (Faxitron HP). Plant were harvested 2 and 26 hours after irradiation, immediatly frozen and stored at -80°C until RNA extraction (Rneasy plant mini Kit, Qiagen) . Experiment was carried in duplicates and a non irradiated control was done for each time.

Gene expression variation in lymphocyte subpopulations in response to low dose of ionizing radiations

Project description:Lymphocytes are the standard cell of a majority of biodosimetry approaches because of their availability and radiosensitivity. But progresses remain to be performed to bring down the minimum threshold of dose sensitivity. To find a lymphocyte subtype able to respond to low doses of ionizing radiation, we examined gene expression variations in different lymphocyte subpopulations using the microarray technology. Blood samples were independently exposed to 0, 0.05 and 0.5 Gray of ionizing radiations. Three and 24 hours after exposure, CD56+, CD4+ and CD8+ cells were negatively isolated. RNA from each condition was competitively hybridized on oligonucleotide microarrays. Keywords: Dose response, stress response, cell type comparison We characterized genes whose expression level was affected following exposure to γ rays by carrying out genome-wide expression profiling on each lymphocyte subpopulation. Total blood samples were collected from 5 healthy male donors. Each sample was splitted into three parts which were exposed respectively to 0 Gy (sham-irradiated), 0.05 Gy and 0.5 Gy of ionizing radiations. Lymphocytes subpopulations were sorted 3 and 24 hours after irradiation. RNA was prepared from each cell type and amplified. The amino-allyl amplified RNAs were labeled and independently hybridized to the array. RNA from MNC obtained from 20 independent healthy donors constituted the reference group for competitive oligonucleotide microarray hybridization. According to our criteria of quality, we considered a total of 114 microarrays

Gene-expression profiling of Saccharomyces cerevisiae irradiated by high-LET radiations

Project description:Ionizing radiations are categorized by linear energy transfer (LET) into low-LET and high-LET. High-LET is considered to have a higher relative biological effectiveness (RBE) than low-LET radiations. However, the details of the effects have not been clearly determined. The aim of this study was to characterize the difference between high-LET and low LET radiations. The global effects of the three types of high-LET radiations (fast neutron, heavy ion (C) and thermal neutron) were compared with the low-LET radiation (gamma ray) using yeast DNA microarrays. Highly induced genes by the three types of high-LET radiations were those genes related to oxidative stress. Oxidative stress was one of the common factors associated with the four types of radiations. Oxidative stress induced by high-LET radiations may be more serious than that induced by gamma rays. Additionally, genes related to protein synthesis and the ubiquitin and proteasome system were detected. This suggests that more protein damages can be induced by high-LET radiation that denatures the proteins in yeast cells. The genes specifically altered by each type of high-LET radiation were also studied. Keywords: stress response

Analysis of gene co-expression networks in skin cells exposed to different doses of ionising radiation at different time points

Project description:Most studies have analysed the effects of high dose radiation such as atomic bomb survivors in Japan, people exposed during the Chernobyl nuclear accident, patients undergoing radiation therapy, uranium miners, etc. However, it has been difficult to measure and assess the risk of cancer in people exposed to lower doses of ionising radiation, such as the people living at high altitudes, who are exposed to more natural background radiation from cosmic rays than people at sea level. We measured the genomic response to X-ray ionising radiation (10 cGy and 100 cGy) in a skin tissue model to compare the effects of low and high dose ionising radiation at different time points. The microarray data was then analysed using state-of-the art “upside-down pyramid” computational systems biology methods to identify genes contributing to the difference in the response to the different radiation doses. The model is reconstructed skin tissue, which is composed of keratinocytes that make up the epidermal layer, and fibroblasts that make up the dermal layer of the skin. Tissues were irradiated with 0, 10, and 100 cGy X-ray radiation. Skin plugs were harvested at 0, 3, 8, and 24 hours post irradiation.

Effects of simulated space radiations on the tomato root proteome

Project description:Plant cultivation on spacecraft or planetary outposts is a promising and actual perspective both for food and bioactive molecules production. To this aim, plants response to ionizing radiations, as an important component of space radiation, must be assessed through on-ground experiments due to the potentially fatal effects on living systems. Hereby we investigated the effects of X-rays and -rays exposure on tomato ‘hairy root’ cultures (HRCs), which represent a solid platform for the production of pharmaceutically relevant molecules, including metabolites and recombinant proteins. In a space application perspective, we used a HRC system previously fortified through the accumulation of anthocyanins, which are known for their anti-oxidant properties. Roots were independently exposed to different photon radiations, namely X-rays (250 kV) and -rays (Co60, 1.25 MeV), both at the intensity of 0.5, 5 and 10 Gy. Molecular changes induced in the proteome of HRCs were investigated by a comparative approach based on Two-Dimensional Difference In-Gel electrophoresis (2D-DIGE) technology and mass spectrometry.
