Human Angiogenin Cytoplasmic Interactomics

Project description:Human Angiogenin (hAng) is a member of the ribonuclease A superfamily and a potent inducer of neovascularization. Herein, protein interactions of hAng in the nucleus and cytoplasm of the human umbilical vein cell line EA.hy926, have been investigated by mass spectroscopy. The first gel-free analysis of hAng immunoprecipitates from the cytoplasmic and nuclear extracts from EA.hy926 cancer human cell line, revealed many statistically significant potential hAng-interacting proteins. The majority of the proteins identified by the mass spectrometry analysis are parts of multiprotein complexes, and some of them are common in nucleus and cytoplasm, including spliceosome, proteasome, and molecular chaperone TRiC/CCT. These complexes are involved in crucial biological pathways, implicating thus the key role of hAng in many vital cellular processes.

We then assessed whether alblation of shuttling activities of SRSF1 affects its nuclear functions.

Transcriptome of cytoplasmic and nuclear small RNAs in innate immune cells

Project description:We isolated RNAs from cytoplasmic or nuclear fraction of dendritic cells, and subject these RNAs for high throughput sequencing. After mapped to mouse genome, sequencing reads (18-30nt) were calculated as the log2 ratio using the normalized TPM (transcripts per million reads) value to determine the expression difference of each sRNA from nuclear and cytoplasmic fractions. We then picked up the superior nucleus-localized sRNAs according to the criteria that abundance of each sRNA was over 20 and x>10y (x: abundance of sRNA from nuclear fraction; y: abundance of sRNAs from cytoplasm fraction).

Cytoplasmic NPM1 maintains the leukemic state through HOX expression [ChIP-seq] 

Project description:NPM1 is the most frequently mutated gene in cytogenetically normal acute myeloid leukemia (AML). In AML cells, NPM1 mutations result in abnormal cytoplasmic localization of the mutant protein (NPM1c). NPM1 mutations are founding genetic lesions, however it is unknown whether NPM1c is required to maintain the leukemic state. Here, we show that loss of NPM1c from the cytoplasm, either through nuclear relocalization or targeted degradation, results in downregulation of homeobox (HOX) genes, and differentiation and cell growth arrest of AML cells. Additionally, we show that HOX downregulation is the earliest transcriptional event following the loss of NPM1c from the cytoplasm, preceding and promoting differentiation. Finally, we show that XPO1 inhibition efficiently relocalizes NPM1c to the nucleus, enables differentiation and prolongs survival of Npm1c+ leukemic mice. We describe an exquisite dependency of NPM1-mutant AML cells on NPM1c, providing the rationale for the use of nuclear export inhibitors in AML with mutated NPM1.
