Project description:Orangutans are an endangered species whose natural habitats are restricted to the Southeast Asian islands of Borneo and Sumatra. For potential species conservation and functional genomics studies, we derived induced pluripotent stem cells (iPSCs) from cryopreserved skin fibroblasts obtained from captive orangutans. We report the gene expression profiles of iPSCs and skin fibroblasts derived from orangtuans. The overall goal was to evaluate gene expression biomarkers of pluripotency in iPSCs and skin fibroblasts derived from PBD-ZSD patients and healthy controls. Dermal fibroblast cultures from 2 orangutans were reprogrammed into iPSCs by transfection with retroviruses designed to express the human OCT4, SOX2, KLF4 and c-MYC cDNA. Fibroblasts and iPSCs were cultured in 1:1 ratio of DMEM:F12 medium supplemented with 20% KOSR (knock-out serum replacement) at 37°C with 5% CO2 until confluence for RNA extraction. The overall goal was to evaluate gene expression biomarkers of pluripotency in iPSCs and original fibroblast cultures.

Project description:We have recently described sustained clinical recovery associated with dampened neuroinflammation and remyelination following transplantation of neural precursor cells (NPCs) derived from human embryonic stem cells (hESCs) in a viral model of the human demyelinating disease multiple sclerosis. In order to test the general applicability of NPC transplantation, we developed a new method for the differentiation and purification of NPCs from pluripotent stem cells (PSCs). Herein, we investigated the therapeutic potential of NPCs generated from human induced pluripotent stem cells (iPSCs) in mice infected with the neurotropic JHM strain of mouse hepatitis virus (JHMV) that induces an immune-mediated demyelinating disease sharing clinical and histologic similarities to the human demyelinating disease multiple sclerosis (MS). JHMV-infected mice intraspinally transplanted with EB (embryoid body)-derived NPCs (EB-NPCs) exhibited decreased accumulation of CD4+ T cells in the central nervous system that was concomitant with reduced demyelination at the site of injection. Dampened neuroinflammation and remyelination was correlated with a transient increase in CD4+FOXP3+ regulatory T cells (Tregs) concentrated within the peripheral lymphatics. Importantly, pathological improvements were limited in comparison to our previous report on hESC-derived NPCs and did not result in significant clinical recovery. Further examination of EB-NPCs by whole genome expression analysis showed them to be significantly different than our previously published cells. These findings highlight an intrinsic disparity in the therapeutic potential of NPCs generated from pluripotent stem cells. Pluripotent stem cells were differentiated to neural precursor cells (NPCs) by two separate methods to study the differences in final cell type. The first method was a 4 factor 9 day directed differentiation. This was applied to WA09 embryonic stem cells (ESCs) as well as two lines of induced pluripotent stem cells (iPSCs) that were developed in our lab HDF51iPSC and HDF69iPSC. The other method was a multistep process that included transition through an embryoid body (EB) stage. This was done with the used on the HDF51iPCS line. For controls we used the original pluripotent stem cell lines (WA09, HDF51iPSC and HDF69iPSC) as well as HDF417 which is a human dermal fibroblast line collected by our lab.

Project description:Although NGLY1 deficiency has been discovered as a result of mutations in the NGLY1 gene, cellular and molecular mechanisms underlying the neurological abnormalities due to NGLY1 malfunction in the brain remain mostly unknown. Using human cerebral organoid (CO) models and systems biology techniques, we uncovered NGLY1 deficiencyinduced alterations at the early stage of cerebral development. Despite the similar vitality and cellular pluripotency of NGLY1-functional and -deficient WA09 hESCs, COs developed from the NGLY1-deficient hESCs had the defective formation of SATB2+ upper-layer neurons and attenuation of STAT3 and HES1 signaling critical for sustaining radial glia. The NGLY1-deficient CO cells, compared with the NGLY1-functional ones, also presented higher vulnerability to multiple stressors. Bulk and single-cell analysis of transcriptomes revealed that NGLY1-deficient COs showed a propensity for premature neuronal differentiation, accompanied by significant downregulation of secretory and transcription factors, including TTR, IGFBP2, and ID4. Supplementing recombinant TTR to NGLY1-deficient CO cells reduced their susceptibility to proteasome inhibition. Ectopic expression of NGLY1 led to IGFBP2 and ID4 upregulation in CO cells developed from NGLY-deficient patientderived induced pluripotent stem cells (iPSCs). Moreover, ID4 expression, STAT3 signaling, and proliferation were enhanced by treatment of recombinant IGFBP2 in CO cells developed from the NGLY1-deficient WA09 hESCs and patient-derived iPSCs. Our findings indicate that NGLY1 could be critical for regulating various stress responses and maintaining neural stem cells (NSCs) in the developing cerebrum. In patients, NGLY1 deficiencyassociated neurological abnormalities, including microcephaly, may be a consequence of aberrations in NSC signaling sustained by NGLY1.

Project description:Hepatocytes generated from human induced pluripotent stem cells (hiPSCs) are unprecedented resources for pharmaceuticals and cell therapy. However, little attention has so far been paid to variations among hiPSC lines in terms of their hepatic differentiation. We developed an improved hepatic differentiation protocol and compared multiple hiPSC lines. This comparison indicated that the hepatic differentiation propensity varies among sibling hiPSC clones derived from the same adult human dermal fibroblasts (aHDFs). In addition, hiPSC clones derived from peripheral blood cells (PB-iPSCs) consistently showed good hepatic differentiation efficiency, whereas many hiPSC clones from adult dermal fibroblasts (aHDF-iPSCs) showed poor hepatic differentiation. However, when we compared hiPSCs from blood and dermal fibroblasts from the same individuals, we found that variations in hepatic differentiation were largely attributable to donor differences, rather than to the types of the original cells. In order to understand the molecular mechanisms underlying the observed variations in hepatic differentiation, we performed microarray analyses of sibling aHDF-iPSC clones, and aHDF- and PB-iPSC clones from the same individuals.
