Project description:Microarrays were used to evaluate the effect of sucrose on gene expression in guard cells. Strips of Arabidopsis leaves were incubated with sucrose or mannitol or no sugars, then the leaves were freeze dried and guard cells were dissected from the leaf strips and analyzed. RNA was extracted from guard cells dissected from leaf strips that had been treated with sucrose or with mannitol or no sugars as controls. Triplicate biological replicates were prepared for the treatments and controls. The RNA was amplified twice with T7 RNA polymerase and hybridized to Affymetrix ATH1 arrays.

Project description:In order to discover new nodes and edges for low CO2-induced stomata closure, we applied hyphenated mass spectrometry (MS)-based metabolomics and proteomics approaches to analyze short-term low CO2 responses in B. napus (canola) guard cells. A total of 411 metabolites were quantified. We observed decreased trends of primary metabolites biosynthesis (e.g., most common amino acids, nucleotides, and sugars), and changes in the levels of osmoregulators such as sucrose, malate, and mannitol showed increased trends at both early and late time points. Unlike under elevated CO2 levels, JA biosynthesis was not altered. Instead, phytohormones that induces stomatal opening, including cytokinins, auxins, GA4, as well as melatonin increased at early time points. This study also highlights the utility of single cell-type metabolomics in discovering and testing new nodes and edges in cellular signaling and metabolic networks.

Project description:We prepared RNA from total leaves and epidermal peels, containing living guard cells (GC) and subsidiary cells (SC) only. In the third sample we blended epidermal peels to mechanically destroy the SC, leading to a pure guard cells fraction. By RNAseq and substraction of GC from SC transcripts, we were able to reliably assign transcripts to the barley SCs and GCs.

Project description:Systemic Acquired Resistance (SAR) as a plant immune response improves immunity of distal tissue after local exposure to a pathogen. Stomata pores on leaf surfaces are formed by guard cells that recognize bacterial pathogens via pattern recognition receptors. We found that Arabidopsis thaliana plants that have been previously exposed to the pathogenic bacteria Pseudomonas syringae pv. tomato DC3000 (Pst) exhibit an altered stomatal response compared to control plants when systemic leaves are later exposed to the bacteria. Reduced stomatal apertures of SAR primed plants lead to decreases in the number of bacteria that enter the apoplastic space of the leaves. To examine how SAR affects molecular processes in guard cells of distal leaves, we collected Arabidopsis guard cell samples and extracted lipids, metabolites and proteins using a 3-in-1 method. Multi-omic results have revealed molecular components of SAR response specific to the functions of guard cells and roles of ROS and fatty acid signaling in guard cell SAR response. Additionally, our results show an increase in palmitic acid and its derivative 9-PAHSA in primed guard cells. Palmitic acid may play a role as an agonist to Flagellin Sensitive 2 (FLS2), which initiates stomatal closure upon perception of bacterial flagella. The improved understanding of how SAR immune signals affect stomatal movement and immunity can aid biotechnology and marker-based breeding of crops for enhanced disease resistance.

Project description:rs05-05_atrazine - effect of atrazine and sucrose on arabidopsis transcriptome - Growth in the presence of sucrose was shown to confer to Arabidopsis thaliana seedlings, under conditions of in vitro culture, a very high level of tolerance to the herbicide atrazine and to other photosynthesis inhibitors (Sulmon et al., 2004). The CATMA investigation will be useful to reveal the gene networks implicated in the mechanisms of xenobiotics tolerance. - Seeds of Arabidopsis thaliana (ecotype Columbia) were surface-sterilized inbayrochlore/ethanol (1:1, v/v), rinsed in absolute ethanol and dried overnight. Germination and growth were carried out under axenic conditions in square Petri dishes. After seeds were sowed, Petri dishes were placed at 4degreeC for 48 h in order to break dormancy and homogenize germination, and then transferred to a control growth chamber at 22degreeC under a 16-h light period regime at 4500 lux for 4 weeks. Growth medium consisted of 0.8% (w/v) agar in 1x Murashige and Skoog (MS) basal salt mix (Sigma, St. Louis, MO, USA) adjusted to pH 5.7. After 4 weeks of cultivation on vertical plates plantlets were transferred on fresh medium complemented or not with atrazine 10 uM and with sucrose 80mM or mannitol 80 mM which were directly added during preparation of agar-MS media prior to sterilisation. Atrazine was sterilized by microfiltration through 0.2 um cellulose acetate filters (Polylabo, Strasbourg, France) and then axenically added to melted agar-MS medium prior to pouring into Petri dishes. Project description:After localized invasion by bacterial pathogens, systemic acquired resistance (SAR) is induced in uninfected plant tissues, resulting in enhanced defense against a broad range of pathogens. Although SAR requires mobilization of signaling molecules via the plant vasculature, the specific mechanisms remain elusive. The lipid transfer protein-defective in induced resistance 1-1 (DIR1-1) was identified in Arabidopsis thaliana by screening for mutants that were defective in SAR. Since then, the structure and lipid-binding properties of DIR1 have been determined, showing that its barrel structure can bind two, long-chain fatty-acid molecules. Several SAR mobile signals, including dehydroabietinal (DA), azelaic acid (AzA), and glycerol-3-phosphate (G3P) are dependent on DIR1 for transport to systemic leaves. Closure of stomata, controlled by guard cells, is a local response to bacteria. Here we demonstrate that stomatal response to pathogens is altered in systemic leaves by SAR, and this guard cell SAR defense requires DIR1. Using a multi-omics approach, we have determined potential SAR signaling mechanisms specific for guard cells in systemic leaves by profiling metabolite, lipid, and protein differences between guard cells in wild type and dir1-1 mutant during SAR. We identified two 18C fatty actyls and two 16C wax esters as putative SAR-related molecules dependent on DIR1. Proteins and metabolites related to amino acid biosynthesis and response to stimulus were altered in guard cells of dir1-1 compared to wild type. Identification of guard cell-specific SAR-related molecules will lead to new avenues of genetic modifications/molecular breeding for disease resistant plants.

These pores are the main route for microbe penetration into leaves. However, plants show a remarkable ability to close the pore when sensing the presence of microbial invaders; a phenomenon recently described as stomatal immunity. This study was initiated to understand the transcriptional regulation of this early plant defense against pathogens.
