Origin of samples: mouse embryos expressing green fluorescent protein (GFP) in neural crest stem cells and later glial cells under the control of the proteolipid protein gene (Plp) promoter. At E9.5 the trunk region was dissected out. At E12, E14, E16, E18 and P0 the sciatic nerve was taken. GFP-expressing cells were isolated by FACS sorting. Three or 4 separate batches of embryos analyzed at each stage.

Project description:A developmental, single-cell transcriptional timecourse analysis was performed on murine pancreas from embryonic days 12, 14, and 17. Whole embryonic murine pancreas tissue was dissociated to single cells, stained with a dead-cell indicator, and subjected to fluorescence activated cell sorting (FACS) to select all live cells. Single cell RNA-sequencing libraries were then subsequently generated for 4,631 cells at E12, 9,028 cells at E14 (comprised of two independent batches), and 4,635 cells at E17. Cells were sequenced at a depth of ~60,000 reads/cell (E14 Batch 1) or ~30,000 reads/cell (E12, E14, and E17 Batch 2). Identification of a novel epithelial progenitor population and multiple novel mesenchymal populations showcases the power of single cell RNA-sequencing to uncover previously uncharacterized cell types, and highlights the dynamic cellular heterogeneity of the developing pancreas.

Project description:We performed gene expression profiling on chicken livers at five important embryonic developmental stages (E7, E12, E14, E17 and E21), which were selected from chicken lines with significant differences in abdominal fat content. Each developmental stage had its unique gene expression pattern and stage-specific differentially expressed genes (DEGs). Furthermore, the three rapid growth periods (E12, E14 and E17) had similar expression patterns. Between the two broiler lines, E17 had the largest number of significantly differentially expressed genes (979), specifically enriched in fatty acid metabolism and biosynthesis, PPAR signaling and glycolysis pathways. Therefore, genome-wide gene expression recapitulated the developmental pattern of chicken embryos, and found the important molecular pathways for hepatic lipid metabolism.

Project description:Background: Studies suggested that mesenchymal stem cells (MSCs) have intrinsic neurogenic potential and can be differentiated into neural stem cell/ neural progenitor cells (NPCs) under specific microenvironment. Manipulation of growth factors is one of the popular method to achieve trans-lineage differentiation of MSCs. Synergistic effect of epidermal growth factor (EGF) and fibroblast growth factor 2 (bFGF) have been widely identified as basic requirement for neural differentiation to take place. Insulin-like growth factor 1 (IGF-1) also known as somatomedin C is an important growth promoting protein during embryonic development which control numerous cellular responses and biological systems. Our recent study has found that the combination of EGF, bFGF and IGF-1 could significantly improved the growth and survivability of MSCs-derived NPCs. Therefore, to understand the genomic mechanism underlying the differentiation in vitro, we have studied the miRNAs profile of MSCs-derived NPCs under IGF-1 influenced conditioned microenvironments. Objectives: To evaluate the effects of IGF-1 in trans-lineage differentiation of MSCs, we have induced MSCs into neural lineage in 3 groups; Group A (positive control) - EGF+bFGF, Group B (Treatment) - EGF+bFGF+IGF-1, and Group C (negative control/ untreated). To unravel the role of regulatory miRNAs involved in the early differentiation, we have performed detailed miRNA profiling for MSCs-derived NPCs at three time intervals (day 1, day 3 and day 5). The data has explored crucial miRNAs involved in early differentiation of MSCs into NPCs. Stage specific MSCs-derived NPCs at Passage 1 were collected for total RNA extraction at three time-points (D1, D3 and D5) and hybridization on Affymetrix miRNA geneChip 2.0 arrays. Each experiment were repeated three times independently (Exp 1, Exp 2 and Exp 3). Group A served as positive control (EGF+bFGF), Group B as treatment (EGF+bFGF+IGF-1) and Group C without growth factor as negative control.

Project description:Recent studies demonstrated that tumor cells with stem cell-like properties can be cultured from human glioblastomas by using conditions that select for the expansion of neural stem cells. We established glioblastoma stem-like (GS-) cell cultures from 9 different glioblastomas, 8 of which generated stably expandable cell lines. Analyzing GS-cell cultures, we discovered two clearly discernable phenotypes. Microarray analysis showed that the 4 GSf cell lines shared expression profiles dominated by genes involved in nervous system development and neuropeptide signaling, while the 5 GSr lines shared expression signatures enriched for extracellular matrix-proteins. Project description:Haematopoietic stem cells (HSCs) are derived early from embryonic precursor cells, such as haemogenic endothelial cells and pre-HSCs. However, the identity of precursor cells remains elusive due to their rareness, transience, and inability to be isolated efficiently. Here we employed potent surface markers to capture the nascent pre-HSCs at 30% purity, as rigorously validated by single-cell-initiated serial transplantation assay. Then we applied single-cell RNA-Seq technique to analyse five populations closely related to HSC formation: endothelial cells, CD45- and CD45+ pre-HSCs in E11 aorta-gonad-mesonephros (AGM) region, and mature HSCs in E12 and E14 foetal liver. In comparison, the pre-HSCs showed unique features in transcriptional machinery, apoptosis, metabolism state, signalling pathway, transcription factor network, and lncRNA expression pattern. Among signalling pathways enriched in pre-HSCs, the mTOR activation was uncovered indispensable for the emergence of HSCs but not haematopoietic progenitors from endothelial cells in vivo. By comparing with proximal populations without HSC potential, the core molecular signature of pre-HSCs was identified. Collectively, our work paves the way for dissection of complex molecular mechanisms regulating the step-wise generation of HSCs in vivo, informing future efforts to engineer HSCs for clinical application.
